Öğe Anti-aging potential and anti-tyrosinase activity of three Hypericum species with focus on phytochemical composition by LC-MS/MS(Elsevier, 2019) Ersoy, Ezgi; Ozkan, Esra Eroglu; Boga, Mehmet; Yilmaz, Mustafa Abdullah; Mat, AfifeHypericum species are one of the most commonly used medicinal plants all over the world and traditional uses of the Hypericum species are mainly characterized by external applications for the treatment of dermatological disorders. The purpose of this study was to investigate the chemical compounds with anti-aging and anti-tyrosinase activities of Hypericum perforatum L., H. calycinum L., and H. confertum Choisy from Turkey and to bring innovation for further studies on developing formulations with Hypericum species for topical applications. In this study, chemical compositions of methanol extracts prepared from flowering aerial parts of the following species collected from Uludag, H. calycinum, H. confertum, and H. perforatum were investigated by LC-MS/MS. Nine metabolites, p-coumaric acid, gallic acid, quinic acid, chlorogenic acid, malic acid, protocatechuic acid, rutin, quercitrin, and isoquercitrin were identified in all analyzed species. To investigate the anti-aging and skin-whitening properties, enzyme inhibition activity of the three species were tested against, collagenase, elastase, hyaluronidase, and tyrosinase enzymes. H. calycinum appeared the most active in all enzyme inhibition activity assays which is mainly due to its rich phenolic content. The methanol extract of H. calycinum showed the highest collagenase, elastase and hyaluronidase inhibitory activities with IC50 values of 51.24 +/- 1.33 mu g/mL, 55.77 +/- 1.32 mu g/mL and 22.17 +/- 0.02 mu g/mL, respectively. Additionally, H. calycinum emerged as the most potential tyrosinase inhibitor among these species with 54.30 +/- 0.49% at 200 mu g/mL concentration. The most abundant compounds determined in H. calycinum methanol extract are, chlorogenic acid (2595.15 +/- 0.179 mu g/g), quercitrin (1805.9 +/- 0.98 mu g/g), quinic acid (885.50 +/- 0.073 mu g/g) and isoquercitrin (346.16 +/- 0.046 mu g/g), respectively. The Salvia species have plenty of usages in traditional medicine, important pharmacological effects and economic value. In this context, this study was con-ducted to evaluate the chemical profile and potential bioactivity of ethyl acetate extracts from 3 different Salvia species; Salvia absconditiflora (Salvia cryptantha-an endemic species), S. sclarea and S. palaestina collected from different localities in Anatolia. Total phenolic-flavonoid contents, antioxidant properties and enzyme inhibition experiments were investigated in tested Salvia species. The phenolic profiles were also determined by liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). The analysis revealed the presence of phenolic compounds in ethyl acetate extracts such as cynaroside, rosmarinic acid, cosmosiin, luteolin, apigenin and acacetin. The highest level of phenolic components was detected in S. absconditiflora with 74.16 mg GAE/g. The best radical scavenging and reducing abilities were found in S. absconditiflora (DPPH: 251.39 mg TE/g; ABTS: 340.16 mg TE/g; CUPRAC: 496.20 mg TE/g; FRAP: 322.94 mg TE/g). Moreover, the inhibition of AChE, BChE, and alpha-amylase has also been studied. The most potent AChE inhibitory ability was recorded in S. sclarea (4.00 mg GALAE/g), followed by S. palaestina (3.38 mg GALAE/g) and S. absconditi-flora (3.01 mg GALAE/g). The extracts exhibited similar amylase inhibitory effects. These results reveal that Salvia species [S. absconditiflora (S. cryptantha), S. sclarea and S. palaestina] confirming their potential can be rich sources of promising bioactive compounds as a starting point for further analysis. In addition, it is thought that this study may be the new report for the construction of helpful databases in terms of shedding light on future research and researchers with the valuable information obtained about these economically important Salvia species. The Euphorbia genus is represented by 105 species and locally named as Sutlegen and Xasil in Turkey. The present study aimed to determine the chemical constituents of E. aleppica, E. eriophora, E. macroclada, E. grisophylla, E. seguieriana subsp. seguieriana, E. craspedia, E. denticulata, E. falcata, and E. fistulosa, and classify them by utilizing the chemometric techniques of principal component analysis (PCA) and hierarchical cluster analysis (HCA). Linoleic acid, 17-tetratriacontane, palmitic acid, and hexatriacontane were the major fatty acids from the gas chromatography-mass spectrometry (GC/MS) analyses. Characterization of 268 constituents of the studied species was achieved by liquid chromatography-ion trap-time-of-flight-mass spectrometry (LC-IT-TOF-MS). Furthermore, a new liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated for the simultaneous quantitative determination of 11 compounds (quinic acid, protocatechuic acid, rutin, hesperidin, eugenol, p-coumaric acid, piceatannol, scopoletin, dl-kavain, chrysophanic acid, and resiniferatoxin) in these species. The developed method was validated for the linearity, limit of detection, limit of quantification, repeatability, and recovery.Öğe Chemical Compositions by Using LC-MS/MS and GC-MS and Biological Activities of Sedum sediforme (Jacq.) Pau(Amer Chemical Soc, 2014) Ertas, Abdulselam; Boga, Mehmet; Yilmaz, Mustafa Abdullah; Yesil, Yeter; Hasimi, Nesrin; Kaya, Meryem Seyda; Temel, HamdiIn this research, the chemical composition and biological activities of various extracts obtained from whole parts of Sedum sediforme (Jacq.) Pau were compared. The amounts of total phenolic and flavonoid components in crude extracts were determined by expression as pyrocatechol and quercetin equivalents, respectively. All of the extracts (petroleum ether, acetone, methanol, and water) obtained from S. sediforme showed strong antioxidant activity in four tested methods. Particularly, the IC50 values of the methanol extract, which was the richest in terms of total phenolic and flavonoid contents, were found to be lower than those of alpha-tocopherol and BHT in beta-carotene bleaching (9.78 +/- 0.06 mu g/mL), DPPH free radical scavenging (9.07 +/- 0.07 mu g/mL), and ABTS cation radical scavenging (5.87 +/- 0.03 mu g/mL) methods. Furthermore, the methanol extract of S. sediforme showed higher inhibition activity than galanthamine against acetyl- and butyryl-cholinesterase enzymes. Also, acetone and methanol extracts exhibited moderate antimicrobial activity against Candida albicans. The main constituents of fatty acid and essential oil were identified as palmitic acid (C16:0) (28.8%) and alpha-selinene (20.4%), respectively, by GC-MS. In the methanol extract of S. sediforme, quercetin, rutin, naringenin, and protocatechuic, p-coumaric, caffeic, and chlorogenic acids were detected and quantified by LC-MS/MS. Results of the current study showed that the methanol extract of S. sediforme may also be used as a food supplement.Öğe Chemical Content by LC-MS/MS, Antiglaucoma, and Antioxidant Activity of Propolis Samples From Different Regions of Türkiye(Wiley, 2024) Izol, Ebubekir; Bursal, Ercan; Yapici, Ismail; Yilmaz, Mustafa Abdullah; Yilmaz, Isa; Gulcin, IlhamiPropolis is a sticky substance produced by bees because of the reaction of beeswax, pollen, and bee enzymes. Particularly, their biological activity and chemical content attract attention. Thus, in this study, the total amount of phenolic and flavonoid substances, Fe3+-Fe2+, Cu2+ (cupric ions reducing activity [CUPRAC]), and Fe3+-TPTZ (ferric ions reducing antioxidant power [FRAP]) reducing, and DPPH center dot and ABTS center dot+ scavenging assays in vitro antioxidant properties of propolis samples obtained from four different provinces of T & uuml;rkiye were determined. In addition, the chemical content of propolis samples was quantitatively determined by LC-MS/MS, and the antiglaucoma property was revealed by hCAII enzyme inhibition. Propolis samples from Ordu presented the highest amounts of total phenolic and flavonoid content (492.3 +/- 5.8 and 96.1 +/- 2.1, respectively) and also highest antioxidant activity (DPPH center dot and ABTS center dot+ IC50 [mu g/mL]: 8.884 +/- 0.84 and 4.589 +/- 0.80, respectively; Fe+3, CUPRAC, and FRAP: 1.051 +/- 0.012, 1.021 +/- 0.008, and 0.957 +/- 0.007 mu g/mL, respectively). hCAII enzyme inhibition was highest in Mu & scedil; propolis (IC50 [mu g/mL]: 8.6) as determined. By LC-MS/MS, 53 different components were screened and 35 bioactive components were determined. According to the results, propolis was found to be a raw material because it contains high concentrations of acacetin, chrysin, caffeic acid, and quinic acid (123.824, 24.759, 47.779, and 16.32 mg analyte/g extract, respectively).Öğe Chemical Profile and Biological Activities of Two Edible Plants: Chemical Investigation and Quantitative Analysis Using Liquid Chromatography Tandem Mass Spectrometry and Gas Chromatography Mass Spectrometry(Taylor & Francis Inc, 2016) Ertas, Abdulselam; Yilmaz, Mustafa Abdullah; Boga, Mehmet; Hasimi, Nesrin; Yesil, Yeter; Goren, Ahmet C.; Temel, HamdiThe objectives of this study were to define the phenolic and fatty acid profiles, anticholinesterase, antioxidant, antimicrobial activities, and total phenolic-flavonoid contents of Lycopsis orientalis and Tragopogon latifolius var. angustifolius which have been used as food source and food supplement in Anatolia and have never been examined before. Rosmarinic and quinic acids (21.11 and 11.46 mg g(-1) extract, respectively) were found to be the most abundant constituents in L. orientalis and T. latifolius var. angustifolius among the studied 27 compounds by liquid chromatography tandem mass spectrometry. In the fatty acid compositions of L. orientalis and T. latifolius var. angustifolius that were determined by gas chromatography mass spectrometry, oleic (29.1%) and palmitic (28.7%) acids were identified as the major components, respectively. The high antioxidant activity of the methanol extract of L. orientalis shows parallelism to its rosmarinic acid content. Besides, this extract showed medium anticholinesterase activity. The results of the present study proves that the L. orientalis might also be used as a food source due to its high phenolic acid content and strong antioxidant property.Öğe Chemical profile by LC-MS/MS, GC/MS and antioxidant activities of the essential oils and crude extracts of two Euphorbia species(Taylor & Francis Ltd, 2015) Ertas, Abdulselam; Yilmaz, Mustafa Abdullah; Firat, MehmetIn this study, it was aimed to investigate the chemical composition and antioxidant activities of two Euphorbia species. The major component of the fatty acid compositions obtained from the petroleum ether extracts was identified as palmitic acid for Euphorbia gaillardotii and Euphorbia macroclada. The main constituents of the essential oils were identified as arachidic acid for E. gaillardotii and tetratetracontane for E. macroclada. Among the 27 studied compounds, hesperidin, rutin, hyperoside and quinic, malic, gallic and tannic acids were found to be the most abundant compounds in the two Euphorbia species. The methanol extracts of E. gaillardotii and E. macroclada showed strong antioxidant activity in all tested methods. Particularly, IC50 values of E. macroclada methanol extract that was the richest in terms of total phenolic-flavonoid contents were found to be lower than alpha-tocopherol and butylated hydroxytoluene in beta-carotene bleaching, 2,2-diphenyl-1-picrylhydrazyl free and ABTS cation radical scavenging methods.Öğe Chemical Profile of Malva Neglecta and Malvella Sherardiana by LC-MS/MS, GC/MS and Their Anticholinesterase, Antimicrobial and Antioxidant Properties With Aflatoxin-Contents(Marmara Univ, Fac Pharmacy, 2017) Hasimi, Nesrin; Ertas, Abdulselam; Oral, Elif Varhan; Yener, Ismail; Alkan, Huseyin; Boga, Mehmet; Yilmaz, Mustafa AbdullahThe purpose of the present work was to determine the phenolic and the fatty acids profiles by LC-MS/MS and GC/MS with their antioxidant, anticholinesterase and antimicrobial activities, and aflatoxin contents of Malva neglecta and Malvella sherardiana. The phenolic contents of M. neglecta and M. sherardiana were determined by LC-MS/MS. Malic and 4-OH benzoic acids were found to be the most abundant compounds in M. neglecta and M. sherardiana, respectively. On the other hand, essential oil and fatty acid compositions were determined by GC/MS analysis. The methanol extracts of the plants showed the highest effect in all antioxidant assays in this study. The methanol extract of M. neglecta showed the highest activity among the petroleum ether, acetone, methanol and water extracts against acetyl-and butyryl-cholinesterase enzymes (53.68% and 63.95% inhibition ratio, respectively). The acetone extracts of M. neglacta and M. sherardiana exhibited moderate activity against C. albicans with 18 and 17 mm inhibion zone diameter, respectively. The results of the present study is also showed that the M. neglecta and M. sherardiana can also be used as a food source due to its high phenolic acid content and good antioxidant property. Also, the samples were aflatoxin free.Öğe Chemical profiling and antioxidant potential of propolis from Pervari, Siirt: discovery of tannic acid and key bioactive compounds(Taylor & Francis Ltd, 2025) Sirma, Erkan; Fidan, Mehmet; Gok, Mesut; Tegin, Ibrahim; Yilmaz, Mustafa Abdullah; Yabalak, ErdalPropolis, a natural product with remarkable therapeutic potential, has gained attention for its antimicrobial, antioxidant, and anti-inflammatory properties. In this study, propolis samples from Sar & imath;yaprak, Kovana & gbreve;z & imath;, and & Ccedil;emikari in Pervari, Siirt province, were analysed comprehensively. The evaluation included wax composition, DPPH and FRAP assays, total phenolic and flavonoid content, and pollen analysis. Phytochemical compounds were identified using GC-MS and LC-MS/MS techniques. Among the regions, & Ccedil;emikari samples exhibited the highest antioxidant activity and phenolic and flavonoid contents, with wax content averaging 60% across all locations. Key compounds identified via RSH-GC/MS included alpha-pinene, beta-pinene, nonadecanal, and nonanal, while GC-MS revealed borneol, sedrenol, and benzyl alcohol. Significant phenolic compounds such as gallic acid, caffeic acid, and tannic acid (reported for the first time in Turkish propolis) were also detected. The high borneol concentration, with its strong anti-inflammatory properties, underscores the unique traits of these samples and their promising antioxidant potential for further research.Öğe Chemical profiling and total thickness-excised wound-healing activity of Pistacia lentiscus L. fruits growing in Algeria(Taylor & Francis As, 2016) Boukeloua, Ahmed; Belkhiri, Abdelmalik; Yilmaz, Mustafa Abdullah; Temel, HamdiThe aim of this study was to determine the chemical characterization and to assess full-thickness-excised wound-healing activity of the oil of Pistacia lentiscus L. fruits grown in Algeria. The fatty acid composition of P. lentiscus L. extracts were analyzed by GC-MS and their total protein amounts were determined by elemental analysis. Total thickness-excised wound-healing activity of hexane extract was carried out on rabbits; comparing oil and wax effects. The results showed that P. lentiscus L. fruits contain considerable levels of oil and protein. Additionally, oleic (46.91 +/- 1.03, 48.34 +/- 0.07) in hexane and cold pressure extracts, respectively, palmitic (25.75 +/- 0.82, 25.44 +/- 0.65) and linoleic (20.59 +/- 0.03, 20.27 +/- 0.81) acids were the major fatty acids. Wound-healing activity was significantly higher in P. lentiscus L. fatty oil compared to the wax, therefore, fats refining is wildly recommended.Öğe A comparative bio-evaluation and chemical profiles of Calendula officinalis L. extracts prepared via different extraction techniques(MDPI AG, 2020) Ak, Guneş; Zengin, Gökhan; Kouadio İbrahime, Sinan; Mahomoodally, Fawzi M.; Picot-Allain, Marie Carene Nancy; Çakır, Oğuz; Bensari, Souheir; Yilmaz, Mustafa Abdullah; Gallo, Monica; Montesano, DomenicoCalendula officinalis L., (marigold), well known for its medicinal properties, has been extensively studied for its therapeutic properties. Nonetheless, as far as the literature could establish, no study has attempted to comparatively assess the biological (antioxidant and enzyme inhibitory potential) of the flowers, leaves, and roots of C. officinalis extracted using conventional (maceration and Soxhlet extraction (SE)) and non-conventional extraction (homogenizer (HAE) and ultrasound (UAE) assisted extraction) techniques. The detailed phytochemical profile of each extract along with the concentration of specific bioactive compounds has also been established. Total phenolic content was highest for the flower extracts while flavonoid content was highest in the leaf extracts. Phytochemical profiling showed that the extraction method influenced the phytochemical composition of the extract. Nicotiflorin was identified in the flower extracts only while amentoflavone occurred only in the roots, inferring that the occurrence of bioactive compounds varies within a plant. The flower extracts showed highest antioxidant potential while the roots extracts were potent inhibitors of cholinesterase and tyrosinase. This study provides valuable data on the influence of extraction techniques on the recovery of bioactive compounds from plants. In an endeavor to scale-up extraction from plant considering the more efficient extraction method is of paramount importance. Moreover, the study highlighted the necessity to thoroughly examine the biological activities of various parts of a plant obtained via different extraction protocols.Öğe Comparative Studies on Phenolic Composition, Antioxidant, Wound Healing and Cytotoxic Activities of Selected Achillea L. Species Growing in Turkey(Mdpi, 2015) Agar, Osman Tuncay; Dikmen, Miris; Ozturk, Nilgun; Yilmaz, Mustafa Abdullah; Temel, Hamdi; Turkmenoglu, Fatma PinarTurkey is one of the most important centers of diversity for the genus Achillea L. in the world. Keeping in mind the immense medicinal importance of phenols, in this study, three species growing in Turkey, A. coarctata Poir. (AC), A. kotschyi Boiss. subsp. kotschyi (AK) and A. lycaonica Boiss. & Heldr. (AL) were evaluated for their phenolic compositions, total phenolic contents (TPC), antioxidant properties, wound healing potencies on NIH-3T3 fibroblasts and cytotoxic effects on MCF-7 human breast cancer cells. Comprehensive LC-MS/MS analysis revealed that AK was distinctively rich in chlorogenic acid, hyperoside, apigenin, hesperidin, rutin, kaempferol and luteolin (2890.6, 987.3, 797.0, 422.5, 188.1, 159.4 and 121.2 mu g analyte/g extract, respectively). The findings exhibited a strong correlation between TPC and both free radical scavenging activity and total antioxidant capacity (TAC). Among studied species, the highest TPC (148.00 mg GAE/g extract) and TAC (2.080 UAE), the strongest radical scavenging (EC50 = 32.63 g/mL), the most prominent wound healing and most abundant cytotoxic activities were observed with AK. The results suggested that AK is a valuable source of flavonoids and chlorogenic acid with important antioxidant, wound healing and cytotoxic activities. These findings warrant further studies to assess the potential of AK as a bioactive source that could be exploited in pharmaceutical, cosmetics and food industries.Öğe Comparison of chemical and biological properties of in vivo and in vitro samples of Salvia siirtica Kahraman, Celep & Dogan extracts prepared with different solvents(Elsevier, 2021) Fidan, Hilal Saruhan; Kilinc, Fatih Mehmet; Yilmaz, Mustafa Abdullah; Akdeniz, Mehmet; Yener, Ismail; Firat, Mehmet; Onay, AhmetThe use of Salvia species among the public and their importance in the scientific world increase due to their numerous pharmacological and biological activities on a daily basis. In this study, the phytochemical contents of different parts (root, branch, leaf, flower, whole) of different specimens of Salvia siirtica (SS) (in vivo and in vitro) extracts prepared with various solvents (petroleumether, chloroformand ethanol) were determined using different techniques (GC-MS and LC-MS/MS) and the results were compared. In addition, biological activities (antioxidant, cytotoxic, anticholinesterase antiurease, antityrosinase, antielastase and anticollagenase) of all samples were determined and compared. The antioxidant potential of the analysed samples was found to be high, and their enzyme activity potential was low. Besides, in vitro SS-TIS (Temporary Immersion System) sample showed high cytotoxic activity (viability% 2.12 +/- 0.06) against MCF-7 (breast cancer) cell line. The results of GC-MS and LC-MS/MS analyses indicated that ferruginol and sugiol could be isolated from the ethanol extract of S. siirtica roots, salvigenin and beta-sitosterol from the chloroform extract of the aerial parts, and phenolic compounds from the ethanol extract of the aerial parts. In addition, the amount of rosmarinic acid, caffeic acid and 12-demethylmulticauline of in vitro samples were found to be higher than those of in vivo samples. Furthermore, all samples, both in vivo and in vitro, contained high levels of rosmarinic acid and beta-sitosterol. The whole chloroform extract (SSWC) could be the source extract for salvigenin (33952.13 mu g/g) and beta-sitosterol (16369.71 mu g/g), and the root ethanol extract (SSRE) for ferruginol (17721.99 mu g/g). As a result, it is understood that the nature of the plant material, the choice of an appropriate solvent and the parts of the material used are quite effective in chemical content. S. siirtica promise potential natural antioxidant agent in food and/or pharmaceutical industry due to its phytochemical content (rosmarinic and caffeic acids, ferruginol, salvigenin) and antioxidant activity. Though Achillea species were chosen as real life samples, the current method is applicable to a wide range of plant species. The developed method was fully validated in terms of linearity, accuracy (recovery), inter-day and intra-day precision (repeatability), limits of detection and quantification (LOD/LOQ) and relative standard uncertainty (U% at 95% confidence level (k = 2)). Reversed-phase ultrahigh performance liquid chromatography was optimized to achive optimum separation for 37 phytochemical compounds and to overcome the suppression effects. MS detection was performed using a triple quadrupole mass spectrometer and negative or positive ionization modes were optimized for each analyte. Multiple reaction monitoring (MRM) was used to quantify the analytes, related molecular ions and transition ions were optimized. Phytochemical screening of ethanol and methanol-chloroform extracts of root and aerial parts of A. coarctata and A. monocephala were performed by using the developed and validated LC-MS/MS method. Root and aerial parts of both species have considerable amounts of certain phenolic-nonphenolic acids (quinic, malic, fumaric, chlorogenic and vanillic acids) and flavonoids (rutin, hesperidin, isoquercitrin, apigetrin, luteolin, apigenin). Additionally, total phenolic and flavonoid amounts, antioxidant (DPPH free radical scavenging assay, ABTS radical cation decolorization assay, beta-carotene lipid peroxidation test system and CUPRAC cupper reduction capacity methods), anticholinesterase, tyrosinase, urease inhibition and cytotoxic activities (on HeLa (Human Cervical Carcinoma Cell Line) of A. coarctata and A. monocephala were also investigated. It has been determined that the studied Achillea species, that are rich in total phenolic-flavonoid and chlorogenic acid contents, have high antioxidant and cytotoxic potential at the same time. According to the results of LC-MS/MS, antioxidant and cytotoxic activity studies, after detailed chemical investigation and toxicity studies on these species, A. coarctata and A. monocephala may be promoted as promising sources of natural agents and used for the development of nutraceuticals or functional food ingredients in future. (Asteraceae) and the assessment of its diverse biological activities. Utilising LC-MS/MS, the phytochemical composition of the 70% methanol extract of Centaurea glastifolia (CG-ME) was thoroughly elucidated, revealing the presence of 30 distinct phytochemical compounds. Notably, major phenolic constituents identified in the extract included quinic acid, chlorogenic acid, luteolin-7-O-glucoside, kaempferol-3-O-glucoside, luteolin, and apigenin-7-O-glucoside. The antioxidant, antibacterial, antiproliferative, and cytotoxic activities of CG-ME were investigated. The CG-ME exhibited a moderate capacity for scavenging DPPH radicals (IC50: 50.05 +/- 1.58 mu g/mL) and FRAP (63.96 +/- 0.39 mg TE/g extract), indicating a moderate level of antioxidant activity. Moreover, CG-ME demonstrated significant antiproliferative effects (GI(50): 1.10 and 1.30 mu g/mL) on cancer cells (C6 and HTC cancer cell lines, respectively) while displaying low cytotoxicity towards normal cells (LC50: >1000 mu g/mL). In terms of antibacterial activity, CG-ME was found to be inactive against tested both Gram-positive and Gram-negative bacterial strains (MIC > 500 mu g/mL). In addition to being consumed as tea and spice, Thymus species are used as folk medicine for respiratory, digestive, skin, circulatory, genital, nervous, visual and urinary diseases. In this study, it was aimed to examine the essential oil and ethanol extract of the root and aerial parts of Thymus pubescens Boiss. et Kotschy ex Celak in the terms of their biological activity and chemical content. The phenolic content of the species was determined by LC-MS/MS, while triterpenoid content, the chemical composition of the essential oil as well as flavour was determined by GC-MS. In addition, their antioxidant and cytotoxic activities, as well as acetyl- (AChE) and butyrylcholinesterase (BChE), urease, tyrosinase, elastase, collagenase, HMG-CoA reductase and angiotensin-converting enzyme (ACE) inhibitory activities were studied. Thymol (53.33%) was determined as the major component in the essential oil, while b-fenchyl alcohol (43.87%) was determined as the major component of the flavour. According to the LC-MS/MS and GC-MS results, it was found that especially the aerial parts of the species have a high amount of rosmarinic acid (3875.76 mg analyte/g extract), quinic acid (2392.55), naringenin (970.39), oleanolic (92785.96) and ursolic (63373.32) acids. The essential oil of T. pubescens species was observed to show high activity in four antioxidant assays, e.g. inhibition of lipid peroxidation, DPPH and ABTS radical scavenging activity as well as CUPRAC, while the ethanol extracts showed moderate antioxidant activity. In enzyme inhibition assays, the aerial parts exerted marked BChE, elastase, and collagenase inhibitory activities (92.43 +/- 1.28%, 42.59 +/- 0.56, and 48.61 +/- 0.39 at 100 mg/mL, respectively). On the other hand, AChE, urease, tyrosinase, HMG-CoA reductase and ACE inhibitory activities of all extracts were from low to moderate levels. In particular, the aerial parts of the species displayed a high cytotoxic effect (vitality%: 6.82 +/- 0.01 at 200 mg/mL) in breast cancer (MCF-7) cell line. Due to its remarkable antioxidant capacity, high content of rosmarinic, oleanolic, and ursolic acids and especially BChE, elastase, and collagenase inhibitory activity, T. pubescens has the potential for using in food supplements, food preservatives, cosmetics, and pharmaceutical industries.(c) 2022 SAAB. Among 59 extracts prepared from the different parts of the mentioned Euphorbia species, E. seguieriana subsp. seguieriana leaf (IC50: 10.41 +/- 0.93 mu g/mL) in lipid peroxidation inhibitory by beta-carotene-linoleic acid assay exhibited the highest activity, while E. grisophylla root in DPPH free radical scavenging (IC50: 0.79 +/- 0.01 mu g/mL), E. grisophylla seed in ABTS cation radical scavenging (IC50: 8.93 +/- 0.02 mu g/mL), E. fistulosa root both in acetyl- and butyryl-cholinesterase inhibitory (53.71 +/- 1.89% and 77.63 +/- 0.60% inhibition, respectively), E. denticulata leaf in urease inhibitory (96.89 +/- 2.00% inhibition), and E. macroclada mixed in tyrosinase inhibitory (86.99 +/- 3.77% inhibition) activities. However in cytotoxic activity studies, E. denticulata against PDF fibroblast cell lines (IC50: 23.04 +/- 0.03 mu g/mL), E. craspedia against HT-29 cancer cell line (IC50: 13.74 +/- 0.02 mu g/mL), E. fistulosa against MCF-7 and DLD-1 cancer cell lines (IC50: 14.04 +/- 0.04 mu g/mL and IC50: 20.23 +/- 0.08 mu g/mL, respectively) indicated quite good activity. According to the LC-MS/MS results, these species were rich in quinic acid, malic acid and tannic acid, rutin, hesperidin and hyperoside. Incidentally, it was found that the E. eriophora possessed very strong irritating potential. A chemometric approach using principal component analysis (PCA) and hierarchical clustering analysis (HCA) techniques were also studied on collected data to reveal the relationship between chemical contents of different parts of these Euphorbia species, and their biological activities, together with regional differences. Additionally, the methanol extract, essential oil and some individual main compounds of Thymus nummularius were tested for antioxidant, anticholinesterase and antimicrobial activities. Besides, HPLC-FLD was used to determine total aflatoxin in the plant material. Among the twenty-seven compounds studied by LC-MS/MS, rosmarinic acid (131,898.9 +/- 6463.0 mu g/g dry-extract) was found to be the most abundant compound in the methanol extract. On the other hand, thymol (60.38%) and terpinyl-acetate (10.49%) were found to be the major compounds of the essential oil. Both the essential oil and the methanol extract of T. nummularius exhibited strong antioxidant activity in the four tested methods. Furthermore, the essential oil showed high inhibitory activity against acetyl-,butyryl-cholinesterase enzymes and very strong antimicrobial activity against all tested microorganisms. Besides, T. nummularius can be used both as rosmarinic acid source and as food supplement due to its non-aflatoxin content and high antioxidant, anticholinesterase and antimicrobial activities. All rights reserved.Öğe Effect of Tunceli Mountain Garlic (Allium tuncelianum) on Rat Heart Tissue Antioxidant Enzyme Levels and Characterization of Phenolic Components(Kahramanmaras Sutcu Imam Univ, 2018) Kutlu, Turkan; Takim, Kasun; Karaaslan, Merve Goksin; Yilmaz, Mustafa AbdullahIn this study, the phenolic compounds of Allium tuncelianum water extracts were determined by LC-MS / MS and HPLC. Wistar albino female rats were used to determine the effect on antioxidant enzyme levels. Five groups of female rats, each containing eight individuals were studied. The first group was control without any treatments. The second group was treated 7,12-DMBA, third, and fourth groups were treated with DMBA + 250, and 500 mg/kg/day of Allium tuncelianum water extracts, fifth group was treated DMBA+ 200 mg/kg of E vitamin (twice a week) for one month, respectively. The rats were feed for one month, slaughtered afterwards and the cardiac tissue was taken for the experiments. Catalase (CAT) and superoxide dismutase (SOD), Malondialdehide (MDA) and Glutatyon (GSH) parameters, which are antioxidant and oxidative stress markers on the heart tissue of Allium tuncelianum water extract were investigated. Result of this study indicated that Allium tuncelianum water extract was poor in phenolic compound diversity but had a very rich content in terms of some other components including pyrocatechol, kainic acid, fumaric acid and malic acid. Antioxidant enzymes (CAT, SOD) levels in heart tissue decreased significantly (p < 0.05) in the oxidative stress groups compared to the control groups (p < 0.05) and Allium tuncelianum water extract showed a decrease in CAT enzyme activity and an increase in SOD enzyme activity relative to the group given 7,12-DMBA. There was also a decrease in MDA levels compared to the 7,12-DMBA group. It can be concluded that Allium tuncelianum water extract can prevent oxidative stress by increasing SOD enzyme activity and reducing MDA levels.
