| dc.description.abstract | Bu çalışmada nitrik oksitin (NO) sürekli fokal serebral iskemide beyin harabiyatine etkileri incelendi. Yetmişdört erkek Wistar albino sıçan altı deney beş kontrol olmak üzere onbir gruba ayrıldı. Deney gruplarına NO yapımını önleyen non selektif bir nitrik oksit sentaz (NOS) inhibitörü N° -nitro-L-arjinin-metil-ester (L-NAME) ve selektif nöronal NOS inhibhörü (7- NI) ile NO donörü L-arginin (L-NAME iskemiden 30 dakika, 7-NI ve L-arjinin ise 60 dakika önce) uygulandı. 7-NI dimetilsülfoksit (DMSO) içinde çözülerek uygulandığı için ayrıca bir DMSO kontrol grubu oluşturuldu. Fokal serebral iskemiden 24 saat sonra optik kiyazma düzeyinden alman enine beyin kesitleri TTC ile boyandı. Kortikal infarkt alan fotografik incelemeyle belirlendi. Genel NOS inhibitörü L-NAME uygulanan sıçanlarda kortikal infarkt alan %47.4 azaldı (p<0.01). spesifik nNOS 'inhibhörü 7-NI ise kortikal infarkt alam %46.5 azalttı P<0.01). NO donörü L-arjinin serebral infarkt alam değiştirmedi ancak, L-NAME ve 7-NI'mn serebroprotektif etkilerini önledi (P<0.01, P<0.01 sırasıyla). îndüklenebilir NOS (iNOS) inhibitörü aminoguanidin uygulanan sıçanlar, enjeksyondan bir saat sonra öldükleri için iNOS 'un setezlediği NO'nun iskemik hasardaki rolü incelenemedi. Sonuçlarımız fokal serebral iskemide nNOS'un aktive olduğu ve NO üreterek beyin hasarına katkıda bulunduğunu göstermekte ve serebral iskemi mekanizmalarında NO nun işe karıştığını ileri süren hipotezi desteklemektedir. | |
| dc.description.abstract | In this study, The effect of nitric oxide (NO) were examined in permanent focal cerebral ischemia Seventy-four male Wistar-albno rats diveded in to fire ischemic controls and six experimental groups. A non selective NO synthase (NOS) inhibitor, Is^-nitro-L-arjinin-metil- ester (L-NAME) and a selective nöronal NOS (nNOS) inhibitor, 7-nitro indazole (7-NI) and NO donor, L-arginine, were administred intraperitoneally 60 minutes before ischemia. Coronal brain sections were taken at the optic chiasm level after 24 hour of permanent focal cerebral ischemia and staned with tetrazoliumchloride and then photographed. Cortical infarct area was determined by image anaysing. Cortical infarct area was reduced significantly by 47.4% in treated with L-NAME when compared with the saline-treated ischemic control group (P<0.01). The nNOS inhibitor 7-NI significantly reduced cortical infarction area by 46.5% when compared with the DMSO- treated ischemic control group. Treatment with NO donor, L-arginine did not altered cortical infarct area, but prevented the cerebroprotective effect of L-NAME and 7-NI ( respectively, P<0.01, P<0.01). The role of iNOS in focal cerebral ischemia was not investigated because of dying of animals after 1 hour of aminoguanidine ainjection. The results suggest that NOS activity after focal cerebral ischemia contributes to tissue damage and support the hypothesis of NO participation in the mechanisms of cerebral ischema. | |
