Özet
Bu çalışmada bazı sıçan dokularında soğuk stresi, adrenomedullin (AdM) ve
metile-adrenomedullin (met-AdM) uygulamalarının total RNA miktarları
üzerine etkilerinin araştırılması amaçlanmış olup, stres hormonlarının
kontrolünde ve salınmasında etkili enzimler ve bunların regülasyonu hakkında
önbilgi verebileceği düşüncesi ile total RNA miktarları saptanmıştır. Deney
esnasında soğuk stresi uygulama grubunda sıçanlar bir hafta boyunca + 10°C
sıcaklığa maruz bırakılmışlar ve AdM ve met- AdM uygulamaları 2000 ng/kg
olarak i.p olarak günde bir kez olmak üzere 1 hafta süre ile yapılmıştır. Toplam
RNA analizinde ise Tümer ve ark., (1997) tarafından kullanılan metottan
yararlanılmıştır. İstatistiksel değerlendirmeler SPSS for Windows Version 12.0
paket program ile yapılmış olup, sonuçlar ortalama ± standart hata ile
verilmiştir. Soğuk stresi uygulama gruplarında total RNA seviyeleri tüm
dokularda kontrolle kıyaslandığında azalmıştır. Kontrol ile soğuk stresi
uygulama grubu arasındaki fark önemli bulunmuştur (p<0.05). AdM uygulama
gruplarında total RNA seviyeleri kontrol ile kıyaslandığında ise azalma
görülmüştür. Total RNA seviyelerindeki bu azalma istatistiksel olarak
önemlidir (p<0.05). Met-AdM uygulama grubunda ise, total RNA seviyeleri
kontrol ile kıyaslandığında azalmıştır ve kontrol ile met-AdM uygulama
grupları arasındaki fark istatistiksel olarak önemlidir (p<0.05). Soğuk stresin
yanı sıra AdM veya Met-AdM uygulanan gruplarda ise total RNA seviyelerinde
istatistiksel olarak önemli bir azalma bulunmuştur (p<0.05). Sonuçlar AdM’in
strese adaptasyonda rol oynayabileceğini göstermektedir. Protein
metilasyonun da anahtar hücresel olaylarda rol oynadığı bilinmekte olup, bu
nedenle metillenmiş adrenomedullin’in stresin düzenlenmesinde muhtemel rolü
olabileceği sonucuna varılmıştır. Metile adrenomedullin’in farklı dokulardaki
yanıtlarını yorumlayabilmek için ise daha ileri düzeyde çalışmalar yapılması
gerekmektedir.
In this study it was aimed to investigate the effects of cold stress, adrenomedullin and methylated adrenomedullin treatments on total RNA levels of some rat tissues. Total RNA levels were determinated for being able to get introductory information about the enzyme levels which play a central role in control and secretion of stress hormons. During experimental process in cold stress treatment, rats were exposed +10 °C temparature for a week. In AdM and met-AdM treatment groups, animals received intraperitoneal (i.p) injection of AdM and met-AdM (2000ng/kg body weight) once a day during a week. Total RNA anyalysis was determinated according to Tümer et.al. (1997). Statical analysis were performed using SPSS version 12.0. Results were shown as mean± standart deviation. In cold stress treatment groups total RNA levels were decreased in all tissue compared to controls. The differences of between control and cold stress treatment groups were significant (p<0.05). In AdM treatment groups total RNA levels were decreased compared to control. The decrease in total RNA levels were found to be statically significant (p<0.05). In met-AdM treatment groups total RNA levels were decreased compared to control. The differences of between control and met-AdM treatment groups were significant (p<0.05). In total RNA level of group which was exposed to cold stress accompanied to AdM or met-AdM a statically significant decrease was found (p<0.05). The results suggested that AdM may play a possible role in adaptation to stress. It is known that methylation of proteins plays important roles in key cellular events therefore we can suggest that methylated adrenomedullin has possible roles in regulation of stress but we need further studies reveal responses of met-AdM in different tissues
