Özet
Amaç: Bu tezin amacı testiste deneysel torsiyon/detorsiyon sonucu oluşturulmuş iskemi/reperfüzyon hasarında menengiç (Pistacia terebinthus L.) ekstraktının koruyucu ve tedavi edici etkisini araştırmaktır. Gereç ve Yöntem: 48 adet Sprague Dawley türü erkek sıçan 6 gruba ayrıldı. Torsiyon-detorsiyon işlemi için hayvanların sağ ve sol testisleri 720 derece döndürülerek 3 saat torsiyon, ardından 3 saat detorsiyon işlemi uygulandı. Sham grubundaki hayvanların skrotumları cerrahi protokol ile açıldı, başka müdahale yapılmadan kapatıldı. Menengiç grubundaki hayvanlara sham grubundan farklı olarak 15 gün boyunca menengiç ekstresi verildi. Torsiyon-detorsiyon grubundaki hayvanlara 3 saat torsiyon, 3 saat detorsiyon işlemi uygulandı, hayvanlar sakrifiye edildi. Menengiç+torsiyon-detorsiyon grubundaki hayvanlara torsiyon-detorsiyon işleminden önceki 15 gün 2 ml/kg, torsiyon-detorsiyon+menengiç grubundaki hayvanlara torsiyon-detorsiyondan sonraki 15 gün 2 ml/kg, menengiç+torsiyon-detorsiyon+menengiç grubundaki hayvanlaraysa torsiyon-detorsiyondan önce ve sonra 15 gün boyunca 2 ml/kg menengiç ekstresi verildi. Deney sonunda hayvanların testisleri alınarak histopatolojik ve immünohistokimyasal değerlendirmeler yapıldı. Bulgular: Sham ve menengiç gruplarındaki hayvanların testis dokularında düzenli seminifer tübüller, bazal lamina üzerinde 5-6 sıralı germinal epitel izlendi. İntertstisyel alandaki kapillerler ile leydig hücrelerinin görünümlerinin normal olduğu görüldü. Torsiyon-detorsiyon grubundaki hayvanlarda seminifer tübüllerde harabiyet, spermatogenik hücrelerde bozulmalar, İnterstisyel alanda hemoraji, ödem ve damarlarda konjesyon izlendi. Menengiç+Torsiyon-detorsiyon grubundaki hayvanların seminifer tübüllerinde kısmen düzelmeler, damarlarda dilatasyon, konjesyon görüldü. Torsiyon-detorsiyon+menengiç ve menengiç+torsiyon-detorsiyon+menengiç gruplarında bazal laminanın sham grubuna benzer özellikler gösterdiği görüldü, damarlarda konjesyon izlendi. Sonuç: Menengiç ekstraktının testiste torsiyon-detorsiyon sonucu gelişen iskemi-reperfüzyon hasarına karşı testis dokusunun rejenerasyonuna katkıda bulunduğu tespit edildi. Tedavi gruplarında Apaf-1 ekspresyonunu baskılayarak hücrelerin yaşamlarına devam etmelerini desteklediği görüldü. Tedavi gruplarında Sox-9 ekspresyonunu arttırarak spermatogenezis sürecinin devam etmesine katkı sağladığı gözlendi.
Aim: The aim of this thesis was to investigate the protective and therapeutic effects of meningic (Pistacia terebinthus L.) extract on ischemia/reperfusion injury induced by experimental torsion/detorsion in the testis. Materials and Methods: 48 male Sprague Dawley rats were divided into 6 groups. For the torsion-detorsion procedure, the right and left testes of the animals were rotated 720 degrees and torsion was performed for 3 hours, followed by detorsion for 3 hours. The scrotums of the animals in the sham group were opened with a surgical protocol and closed without any further intervention. The animals in the meningic group were given meningic extract for 15 days different from the sham group. The animals in the torsion-detorsion group were subjected to torsion for 3 hours and detorsion for 3 hours, and the animals were sacrificed. The animals in the meningic+torsion-detorsion group received 2 ml/kg of meningic extract for 15 days before torsion-detorsion, the animals in the torsion-detorsion+meningic group received 2 ml/kg of meningic extract for 15 days after torsion-detorsion, and the animals in the meningic+torsion-detorsion+meningic group received 2 ml/kg of meningic extract for 15 days before and after torsion-detorsion. At the end of the experiment, testes of the animals were removed and histopathologic and immunohistochemical evaluations were performed. Results: In the testicular tissues of animals in sham and meningic groups, regular seminiferous tubules and 5-6 rows of germinal epithelium were observed on the basal lamina. Capillaries and leydig cells in the interstitial area were normal. In the torsion-detorsion group, destruction of seminiferous tubules, deterioration in spermatogenic cells, hemorrhage, edema and congestion in the interstitial area were observed. Partial recovery in seminiferous tubules, dilatation and congestion were observed in the seminiferous tubules of animals in Menengiç+Torsion-detorsion group. In torsion-detorsion+menengic and menengic+torsion-detorsion+menengic groups, basal laminae showed similar characteristics to the sham group and congestion was observed in the vessels. Conclusion: Menengiç extract was found to contribute to the regeneration of testicular tissue against ischemia-reperfusion injury caused by torsion-detorsion in the testis. It was observed that it supported the survival of cells by suppressing Apaf-1 expression in the treatment groups. It was observed that it contributed to the continuation of spermatogenesis process by increasing Sox-9 expression in the treatment groups.
