dc.description.abstract	Çalısmamızda, streptozotosin (STZ) ile ratlarda olusturulan diyabetin neden olduğu böbrek hasarını önlemede octreotide ve epidermal growth faktör'ün (EGF) etkilerini ısık ve transmisyon elektron mikroskobu (TEM) düzeyinde incelenmesi amaçlandı. Eriskin erkek 40 adet Sprague-Dawley cinsi rat; kontrol, diyabetik, diyabetik+octreotide, diyabetik+EGF ve diyabetik+octreotide+EGF olmak üzere 5 esit gruba bölündü. Deneyin baslamasından bir ay sonra sakrifiye edilen ratların böbrek dokuları ısık ve elektron mikroskobunda incelendi. Diyabetik gruba ait bazı tubül lümenlerinde protein ve epitel hücre sitoplazmasında PAS (+) boyanan farklı çaplarda glikojen birikimi saptandı. Mezengial matriks artısı, Bowman mesafesinde daralma yanı sıra pedisel ve kapiller fenestratalarda obliterasyon, bazal membranlarda kalınlasma ve mikrovillus transformasyonu izlendi. Octreotide grubunda; mezengial matriks artısı, proksimal tubül hücreleri arasında ayrısma ve glomerular bazal membran kalınlasması tesbit edildi. EGF grubunda podositler tarafından fagosite edilmis apoptotik cisimcikler, mezengiumda elektron yoğun birikimler izlendi. Octreotide ve EGF grubunda kontrol grubuna benzer görüntüler saptandı. Sonuç olarak STZ ile olusturulan diyabetin neden olduğu böbrek hasarını önlemede octreotide ve EGF kombinasyonunun etkin olduğu kanaatine varıldı. dc.description.abstract	In this study we evaluated preventive effects of octreotide and epidermal growth factor (EGF) on the development of diabetic nephropathy in STZ-diabetic rats at the transmission electron microscopy (TEM) and light microscopic level. Male, adult 40 Sprague-Dawley used in this study divided into 5 equal groups as control, diabetic, diabetic+octreotide, diabetic+EGF, diabetic+octreotide+EGF. All rats were sacrificed after 1 month and their kidney tissues were examined in light and electron microscopy. In diabetic group protein deposition in some of the tubular lumens and PAS (+) glycogen deposition with various diameters as vacuole were determined in cytoplasm of tubules epithelium. Increasment in mesangial matrix, narrowing in Bowmann space, obliteration of capillary and pedicell fenestratas, thickening of glomerular basement membran and transformation of microvillus were seen. In octreotide group; increased in mesangial matrix, thickening of glomerular basement membran and decomposed between proximal tubules cells were established. In EGF group; electrodens deposition in mesengium and apoptotic material that were phagocyted by podocytes were seen. In octreotide acetate and EGF group; view were similar to control group In conclusion; combination of octreotide and EGF seem to be more effective in prevention of renal injury in STZ-induced diabetic rats.
