Öğe The effect of benzalkonium chloride on rabbit nasal mucosa in vivo(Springer-Verlag, 2002) Cüreoglu, S; Akkus, M; Osma, Ü; Yaldiz, M; Oktay, F; Can, B; Güven, CIn this study we investigated the histopathological changes of nasal mucosa after the use of topical benzalkonium-chloride solution. In the study, 28 male New Zealand white rabbits were used. The animals were divided into two groups. In the first group, 50 mul of 0.001% benzalkonium-chloride solution was sprayed into the right nostril and 50 mul of 0.9% saline into the left twice daily for 14 days. In the second group, the same solutions were applied twice daily for 28 days. All of the biopsies from each nasal cavity were processed for electron microscopy. In the nostrils treated with 0.001% benzalkonium chloride, there were areas with obvious squamous cell metaplasia. Some cells showed broken cristae of the mitochondria. Degenerative changes in the supportive and olfactory cells, deformation of nuclei and the increase of heterochromatin were observed. The present investigation has shown that the application of topical benzalkonium chloride does induce degenerative morphologic changes in the nasal mucosa in rabbits in vivo.Öğe Immunohistochemical analysis of CD31, CD36, and CD44 antigens in human omentum(Saudi Med J, 2004) Yildirim, A; Akkus, M; Nergiz, Y; Yuruker, SObjective: Milky spots in the human omental tissue are known to be consisting of lymphocytes, macrophages and mast cells. Our goal was to evaluate the relationship of lymphoid cells and macrophages with vasculature and stromal components. Methods: In this study we examined the biopsy specimens obtained from the adult patients whom were operated for different purposes in the General Surgery Department of Dicle University Hospital, Ankara, Turkey. We used CD31 as an endothelial cell marker, CD36 which is known to react with microvascular endothelium and adipocytes, and CD44 which is a hyaluronic acid receptor using an indirect immunoperoxidase technique. Results: We observed that CD31 was mainly reactive with vascular endothelial cells and platelets, CD36 was reactive with microvascular endothelium and adipocytes and CD44 was mainly expressed by the endothelial cells of high endothelial venules, fibroblasts in stromal compartments and by large mononuclear cells. Conclusion: We determined the structural and immunophenotypic features of omental lymphoid tissue components stressing vascular and stromal elements, and we briefly discussed the significance of the expression of these molecules in the determined locations.Öğe Intravesical oxybutynin application: Ultrastructural effects on bladder epithelium.(Mbr Press Inc, 1999) Ersay, A; Ketani, MA; Nergiz, Y; Demirtas, OC; Akkus, M; Otludil, BIntravesically applied oxybutynin, rapidly absorbs into the bloodstream, additional to profound local effect. Currently morphologic effects of oxybutynin on local bladder tissue relatively well established at light microscopic level, but not ultrastructural level. Thirty New Zealand White female rabbits were catheterized daily and intravesical instillation were performed with whether 1mg/kg oxybutynin solution or saline for 30 days. The local effects of the drug on bladder epithelium at electron microscopic level were examined comparing with saline administration. Urinary tract infection (UTI) incidence were similar in both saline and oxybutynin groups (9 vs. 10 of 15 animals respectively) (p>0.05). Interestingly, in 4 of 5 animals that received oxybutynin and never had UTI during the study, separation of zonula occludens intercellular junction was determinated by Jeol electron microscope. This observation can explain rapid absorption of oxybutynin through the bladder into the blood stream. Oxybutynin can damage bladder surface epithelium at ultrastructural level. This effect may lead to increase absorption of it, but not result in higher incidence of bacterial infection.Öğe Mucinous tumors of the ovary(I R O G Canada, Inc, 2001) Yaldiz, M; Uzunlar, AK; Kilinç, N; Yalinkaya, A; Akkus, M; Tunik, SOvarian mucinous tumors stem from ovarian surface epithelium and are divided into benign, borderline and malignant. If is difficult to differentiate borderline and malignant mucinous tumors. Thirty-eight cases of ovarian mucinous tumors which were diagnosed at the Pathology Department of Dicle University Medical Faculty were reviewed. Of these, 18 (47.3%), six (15.7%) and 14 (36.8%) were benign, borderline and malignant. respectively. The patients' ages ranged from 18 to 67 (average 44.5) years. Bilaterality was detected in 1/18 (5.5%). 0/6 and 4/14 (28.5%) of benign, borderline and malignant mucinous tumors, respectively. Mean tumor size was 26.4 cm. Microscopically, there was no stratification in the benign tumors. The borderline tumors had papillary infoldings and 2-3 layers of atypical epithelial cells but no invasion of the stroma. Malignant tumors had four or more layers of atypical epithelial cells and stromal invasion.Öğe Replacement of circumflex branch of left coronary artery originating from the right aortic sinus(Springer France, 2001) Karadede, A; Tacar, O; Akkus, MReplacement of the circumflex branch of the left coronary artery is extremely rare. We conducted a retrospective study to determine the incidence of this anomaly of circumflex branch of the left coronary artery. Two such patients were identified from 850 adults undergoing cardiac catheterization procedures. The circumflex branch of the left coronary artery was replaced by a vessel arising from the right aortic sinus. The course of this vessel was retroaortic. A knowledge of this anomaly is important for coronary artery bypass surgery.Öğe Structural and immunophenotypic characterization of high endothelial venules in rat and human tissue(Saudi Med J, 2004) Akkus, M; Yuruker, S; Dagdeviren, A; Asan, EObjective: To present additional data on high endothelial venule (HEV) structure and immunophenotype. Methods: We used the zinc iodide-osmium tetroxide technique (ZIO), which is a metallophilic fixation and staining technique to examine HEVs at light and electron microscopic levels as this technique was previously reported to be reactive with cells in HEVs. Tonsils and lymph nodes were obtained from the Surgery and Otorhinolaryngology Departments, Hacettepe University Hospital, Ankara, Turkey during 2002 and 2003. An indirect immunohistochemical technique was used to examine frozen human tissue samples. Results: Organelle rich high endothelial cells, sheet-like processes of pericytes surrounding HEVs, structural relation of pericyte processes with fibroblastic reticular cells, an unusual multivesicular body-like organelle within high endothelial cells were presented. Expression of a large panel of defined and yet non-defined antigens on HEVs are also presented using an indirect immunoperoxidase technique. Conclusion: Presence of some of these antigens on HEVs was previously reported while no previous report is available for others.
