Öğe 460 bazal hücre karsinomlu olgunun histopatolojik değerlendirilmesi(1999) Yaldız, Mehmet; Yılmaz, Fahri; Kılınç, Nihal; Büyükbayram, Hüseyin; Arslan, Adem; Uzunlar, Ali KemalBazal hücreli karsinomlar derinin en yaygın malign tümörüdür. Bu çalışmada 460 bazal hücreli karsinomun histopatotolojik özellikleri gözden geçirildi ve histopatolojik alt türleri ile cerrahi sınırlarda tümör pozitifliği, lenfosit infiltrasyonu ve ülserasyon varlığı araştırıldı. Olgularımız 6 ana histolojik alt tür açısından değerlendirildi; 210'u (%45.6) nodüler, 93'ü (%20.2) mikst, 62'si (%13.5) infiltratif, 45'i (%9.8) süperfisial, 43'ü (%9.4) mikronodüler, 7'si (%1.5) morfeiform tipti. Materials and methods : Thirty male Sprague-Dawley rats were used in the present study, which were divided into three groups (sham group, n = 10, and two experimental groups, n = 10 for each group). Rats in the experimental group were exposed to 100 and 500 mu T ELF-MF (2 h/day, 7 days/week, for 10 months) corresponding to exposure levels that are considered safe for humans. The same experimental procedures were applied to the sham group, but the ELF generator was turned off. Tissues from the testes were immunohistochemically stained for active (cleaved) caspase-3 in order to measure the apoptotic index by a semi-quantitative scoring system. The levels of catalase (CAT), malondialdehyde (MDA), myeloperoxidase (MPO), total antioxidative capacity (TAC), total oxidant status (TOS), and oxidative stress index (OSI) were also measured. Additionally, epididymal sperm count and sperm morphology was evaluated. Results : There were no signifi cant diff erences in the reproductive and oxidative stress parameters between the sham group and the exposed groups (p > 0.05). While no diff erence was observed between the fi nal apoptosis score of the sham and the 100 mu T ELF-MF group (p > 0.05), the fi nal apoptosis score was higher in the 500 mu T ELF-MF exposure group than in the sham group (p < 0.05). Conclusion : Long-term exposure to 100 mu T and 500 mu T ELF-MF did not aff ect oxidative or antioxidative processes, lipid peroxidation, or reproductive components such as sperm count and morphology in testes tissue of rats. However, long-term exposure to 500 mu T ELF-MF did aff ect active-caspase-3 activity, which is a well-known apoptotic indicator.Öğe A case of giant benign localized fibrous tumor of the pleura(2000) Uzunlar, Ali Kemal; Yılmaz, Fahri; Yaldız, Mehmet; Balcı, Akın E.; Özercan, İbrahim H.A 57-year-old man presented with progressively increasing shortness of breath. A chest roentgenogram showed the presence of a giant mass and magnetic resonance of the chest confirmed the mass with an inhomogeneous density in the right hemithorax. The patient underwent a thoracotomy. A tumor was seen to have arisen from the parietal pleura. Size of the tumor was 22x19x15 cm and weighed 2650 g. It was successfully resected. Olgularımızın 21'i kadın 36'sı erkek olup erkek kadın oranı 1.71'di. Yaş ortalaması 36.73 idi ve en çok olgu ikinci ve beşinci dekatlarda izlendi. Daumas-Duport yöntemine göre olguların %7'si derece I, %16'sı derece II, %32'si derece III ve %45'i derece IV'tü. Dünya Sağlık Örgütü (DSÖ) sınıflandırmasına göre ise olguların %23'ü astrositom, %32'si anaplastik astrositom ve %45'i glioblastom idi. The aim of the study was to investigate the long-term effect of 900 MHz RFR and ELF-MF exposure on nasal mucosa and nose skin of rats. In this study two different exposure systems were used to get information about the effects of RFR emitted from mobile phones and ELF-MF on nose. In the first part of the study Twenty one Wistar Albino adult male rats (sham: 7, exposure: 14) were used to determine the effects of 900 MHz RFR on nose. For the exposure group, rat heads were exposed to the 900 MHz RFR 2 h per day (7 days in a week) for 10 months. For the sham group, same process was applied to the rats, except that the generator was turned off In the second part of the study 30 male Sprague-Dawley rats (Mona: 10, first exposure: 10, second exposure: 10) were used to determine the effects of 100 mu T and 500 mu T extremely low frequency magnetic field (ELF-MF) which complies to the safety standards for public and occupational exposure. The rats in the exposure groups of this part of the study were exposed to 100 mu T and 500 mu T ELF-MF for 2 hours per day (7 days in a week) during 10 months in a Plexiglas cage. For the sham group, the same experimental process was applied to the rats but ELF-MF generator was turned off The results of the present study showed that 10 months of exposures of mobile phones and ELF-MF do not affect the mucous membrane and skin of nose. In conclusion, radiofrequency and ELF-MF used in this study do not affect nasal skin and mucosa of rats exposed to 900 MHz RFR, 100 mu T and 500 mu T ELF-MF.Öğe Effect of Mobile Phone Exposure on Apoptotic Glial Cells and Status of Oxidative Stress in Rat Brain(Taylor & Francis Inc, 2009) Dasdag, Suleyman; Akdag, M. The study carried out on 31 Wistar Albino adult male rats. The rat heads in a carousel exposed to 900MHz microwave. For the study group (n:14), rats exposed to the radiation 2 h per day (7 days in a week) for 10 months. For the sham group (n: 7), rats were placed into the carousel and the same procedure was applied except that the generator was turned off. For the cage control (n: 10), nothing applied to rats in this group. In this study, rats were euthanized after 10 months of exposure periods and brains were removed. Brain tissues were immunohistochemically stained for the active (cleaved) caspase-3, which is a well-known apoptosis marker, and p53. The expression of the proteins was evaluated by a semi-quantitative scoring system. However, total antioxidative capacity (TAC), catalase, total oxidant status (TOS), and oxidative stress index were measured in rat brain. Final score for apoptosis in the exposed group was significantly lower than the sham (p < 0.001) and the cage control groups (p < 0.01). p53 was not significantly changed by the exposure (p > 0.05). The total antioxidant capacity and catalase in the experimental group was found higher than that in the sham group (p < 0.001, p < 0.05). In terms of the TOS and oxidative stress index, there was no statistically significant difference between exposure and sham groups (p > 0.05). In conclusion, the final score for apoptosis, total antioxidant capacity and catalase in rat brain might be altered by 900MHz radiation produced by a generator to represent exposure of global systems for mobile communication (GSM) cellular phones.Öğe Effects of Extremely Low-Frequency Magnetic Field on Caspase Activities and Oxidative Stress Values in Rat Brain(Humana Press Inc, 2010) Akdag, Mehmet Zulkuf; Dasdag, Suleyman; Ulukaya, Engin; Uzunlar, Ali Kemal; Kurt, Mustafa Ayberk; Taskin, AbdullahThis study was aimed to investigate the effect of extremely low-frequency magnetic field (ELF-MF) on apoptosis and oxidative stress values in the brain of rat. Final score of apoptosis and MPO activity were not significantly different between the groups. CAT activity decreased in both exposure groups (p<0.05), while TAC was found to be lower in ELF 500 group than those in ELF-100 and sham groups (p<0.05). MDA, TOS, and OSI values were found to be higher in ELF-500 group than those in ELF-100 and sham groups (p<0.05). In conclusion, apoptosis was not changed by long-term ELF-MF exposure, while both 100 and 500 mu T ELF-MF exposure induced toxic effect in the rat brain by increasing oxidative stress and diminishing antioxidant defense system.Öğe Effects of specific inhibition of cyclooxygenease-2 on kidney in bilateral adrenalectomized rats(Springer, 2007) Ozturk, Hulya; Ozturk, Hayrettin; Gedik, Senol; Uzunlar, Ali Kemal; Ketani, AydinIn the kidney, prostaglandins represent important physiological modulators of renal hemodynamics and salt and water homeostasis. In this experimental study of bilaterally adrenalectomized (ADX) rats, we aimed to investigate whether the administration of selective (celecoxib) inhibitor of COX-2 would alter the morphological and functional changes in rat kidney tissue. Twenty-one male Sprague-Dawley rats weighing 225-250 g were used. The animals were divided into three groups. Group 1 rats (Sham-control, n = 7) did not receive any treatment. In group 2 rats (ADX/Untreated, n = 7), bilateral ADX was performed via a single dorsal incision. In group 3 (ADX/COX-2) rats, the same operation was performed as described for group 2 and then the COX-2 inhibitor celecoxib was administered by gavage for a period of 7 days. On the 7th day of the study, renal function was assessed by measurements of blood urea nitrogen (BUN) and serum creatinine levels. Biopsies were obtained from the remaining left kidneys before killing the rats. There was no significant difference in the BUN and creatinine values between the groups. In ADX/Untreated group, capillary congestion in glomerule, inflammation, hemorrhage and congestion in intertubular area, and cytoplasmic vacuolation in renal tubules was observed. Mild damage was observed in the ADX/COX-2 group. The number of macrophages was significantly decreased in ADX/COX-2 group when compared to ADX/Untreated group (P < 0.0001). Our study indicates that celecoxib may be an important factor affecting renal morphological changes after the bilaterally ADX.Öğe Experimental study on effects of deferoxamine mesilate in ameliorating cisplatin-induced nephrotoxicity(Akademiai Kiado Rt., 2002) Özdemir, Enver; Dokucu, Ali İhsan; Uzunlar, Ali Kemal; Ece, Aydın; Yaldız, Mehmet Sadık; Öztürk, Hayrettin; 0000-0001-6764-8336Purpose: Cisplatin (CCDP), an indispensable agent of several chemotherapy protocols, has serious dose limiting side effects, including nephrotoxicity. In this experimental study, we used deferoxamine mesilate (DFO), an iron chelating agent, to ameliorate cisplatin-induced nephrotoxicity. Materials and methods: Sixty adult male bulb-c mice were divided in 6 equal groups. Group 1 received distilled water, group 2 received 100 mg/kg DFO, group 3 received 0.9 mg/kg CCDP, group 4 received 100 mg/kg DFO one hour before 0.9 mg/kg CCDP, group 5 received 1.8 mg/kg CCDP, and group 6 received 200 mg/kg DFO one hour before 1.8 mg/kg CCDP transperitoneally for 10 days. The next day, blood and urine samples were obtained, and all the animals were sacrificed, the kidneys and testes were removed, and histopathologic and biochemical analyses were performed. Results: Low-dose and high-dose CCDP treated mice had significantly more extensive proximal tubular degeneration (p < 0.001) when compared to control animals. Moreover, these changes were significantly less extensive in the mice taking DFO than mice taking CCDP. DFO showed no effect on cisplatin induced testicular histopathology. The cisplatin administration significantly increased the serum urea and plasma creatinin concentrations, and DFO administration prior to CCDP significantly decreased serum urea and plasma creatinin concentrations. Conclusion: Our findings suggest that DFO administration may be safe and useful for ameliorating cisplatin-induced nephrotoxicity.Öğe Fasioliazis; nadir bir kronik kolesistit nedeni(2009) Uzunlar, Ali KemalFasioliazis, fasiola hepaticanın neden olduğu zoonotik bir infestasyondur. 27 yaşında bayan bir hastada Fasciola hepaticanın neden olduğu kronik kolesistit olgusu sunuldu. Tanı, safra kesesi içinde makroskopik olarak izlenen parazitler ve serolojik incelemeyle kondu. Fasioliazisin nadiren de olsa kronik kolesistite neden olabileceği bilinmesi gerekir.Öğe Grading of the ovarian serous cystadencarcinomas, and its correlation with BRCA1 and p53 immunoreactivity(Ortadogu Ad Pres & Publ Co, 2007) Guenes, Nedim; Yilmaz, Fahri; Uzunlar, Ali KemalObjective: Ovarian cancer is a gynecological malignancy with the higest mortality. The surface epithelial tumors of the ovary are the most common cancer types. The purpose of this study was to grade the ovarian serous cystadenocarcinomas and to evaluate BRCA1 and p53 proteins immunohistochemically. Material and Methods: Fifty-eight cases diagnosed as serous cystadenocarcinomas of the ovary were selected for this study. The cases were graded according to two-tier grading system, FIGO and Shimizu-Silverberg grading system and the results were compared. The BRCA1 and p53 proteins were studied immunohistochemically. Results: The cases were distributed as follows: according to the two-tier grading system low grade 12 (21%) cases and high grade 46 (79%) cases; according to the FIGO grading system grade 19 (16%) cases, grade B 29 (50%) cases, and grade 11120 (34%) cases and according to the Shimizu-Silverberg grading system grade 111 (19%) cases, grade II 29 (50%) cases, and grade 11118 (31%) cases. Positive p53 staining was detected in 57% (33/58) and BRCA1 was detected in 67% (39/58) of the cases. The relationship between p53 and BRCA1 proteins and histopathologic grade was analyzed. High grade ovarian serous carcinomas had a higher rate of p53 expression (65%) compared to low grade (25%) tumors. BRCA1 immunstaining was detected in 92% (11/12) of low grade and 61% (28/46) of high grade tumors according to two-tier grading system. Conclusion: There is a good correlation between the two-tier grading system and the FIGO and the Shimizu-Silverberg grading systems. In addition, the application of the two-tier grading system for ovarian serous carcinomas is easy because this system is based on easily understood two criteria and includes two categories (low grade, high grade). In contrast to p53, high grade tumors had a lower rate of BRCA1 expression (61%) compared to low grade (92%) tumors. Objective: We tried to integrate cervical cancer screening programme with a family planning service in a family planning clinic. Design: Volunteer women, who can speak the local language, were assigned to educate residential women on cervical cancer and to refer them to a family planning clinic. All nurses working in the family planning clinic were trained on how to perform cervical smear. Setting: The study was conducted in Huzurevleri district of Diyarbakir-Turkey. Participants: The Pap test results of 503 women who gave informed consent and attended the family planning clinic for cervical smear test were the participants of the study. Women's practices and previous Pap test history were also discussed. Main Outcome Measure: To examine the effect of factors influencing Pap test history frequency tabulates, chi-square and logistic regression analyses were performed. Results: Within one year, 503 Pap test were investigated. Although 361 women (71.8%) attended clinic previously, only 37 women (7.4%) had a Pap test. Illiteracy and history of induced abortion were the factors affecting Pap test usage. Adjusted odds ratio for illiterate women, who had not had a Pap test before, was 2.80 (95% CI: 1.3-6.3) and for women who had never induced abortion was 3.88 (95% CI: 1.3-12.0). Conclusion: Integration of cervical cancer screening with family planning services may avoid missed opportunities. Especially illiterate women should be reached because of their risks.Öğe Inguinal endometriosis: A report of two cases and a review of the literature(2000) Uzunlar, Ali Kemal; Yılmaz, Fahri; Yaldız, Mehmet; Özaydın, Mehmet; Kaya, Özden[Abstract Not Available]Öğe Kongenital gingival granüler hücreli tümör: Olgu sunumu(2006) Uzunlar, Ali KemalÖZET: Amaç: Bu çalışmada, nadir görülen ağız içi yerleşimli bir konjenital gingival granüler hücreli tümör (KGGHT) olgusu sunulması amaçlandı. Olgu: Yenidoğan bir kız bebekte, fizik muayenede, ağız içinden dışarıya taşan 3 cm çapında kitle saptandı. Kitle, aspirasyon pnömoni riski ve beslenme sorunları nedeniyle operasyonla çıkarıldı. Makroskopik olarak saplı düzgün yüzeyli tümör izlendi. Mikroskopik olarak; tümör oval nükleuslu ince granüler geniş eozinofilik stoplazmalı, hücresel atipi ve mitoz içermeyen hücrelerden oluşmaktaydı. Beş aylık takip sonunda herhangi bir komplikasyon veya rekürrens izlenmedi. Sonuç: KGGHT, klinik olarak kimi lezyonlarla karışabilmesine rağmen, patolojik olarak tanınmasında herhangi bir zorluk bulunmamaktadır.
