Öğe Analysis of bioactive compounds using LC-ESI-MS/MS, cytotoxic, antimicrobial effects, and enzyme activities from Cyclotrichium origanifolium(Wiley, 2023) Aktepe, Necmettin; Baran, Ayse; Atalar, Mehmet Nuri; Baran, Mehmet Firat; Keskin, Cumali; Taskin, Abdullah; Yavuz, OemerCyclotrichium origanifolium is a medicinal plant belonging to the Lamiaceae family. In this study, phenolic content analysis, antimicrobial effects, and cytotoxic effects of extracts of C. origanifolium were investigated. In the extracts, phenolic compound analysis by the liquid chromatography-electrospray ionization-tandem mass spectrometry method, antimicrobial effect by the minimum inhibition concentration method, and cytotoxic effect on human dermal fibroblasts (HDF), glioblastoma cell (U87), ovarian adenocarcinoma cell (Skov-3), and human colorectal adenocarcinoma cell (CaCo-2) cancer cell lines were investigated. Cytotoxicity analyses were performed by the MTT method. In addition, the GST and AChE enzyme activities of the extracts were also measured. Around 18 compounds were detected in both the methanol and ethanol extract. It was found that the best antimicrobial effect on Gram-negative Pseudomonas aeruginosa was on methanol extract, while the ethanol extract was on Candida albicans fungus (respectively, 2.50 mg/ml, 5.0 mu g/ml). A 500 mu g/ml of methanol extract has been shown to have cytotoxic activity high effect on HDF cells. GST and AChE activity were found to decrease in a concentration-dependent manner.Öğe Effects of Extremely Low-Frequency Magnetic Field on Caspase Activities and Oxidative Stress Values in Rat Brain(Humana Press Inc, 2010) Akdag, Mehmet Zulkuf; Dasdag, Suleyman; Ulukaya, Engin; Uzunlar, Ali Kemal; Kurt, Mustafa Ayberk; Taskin, AbdullahThis study was aimed to investigate the effect of extremely low-frequency magnetic field (ELF-MF) on apoptosis and oxidative stress values in the brain of rat. Rats were exposed to 100 and 500 mu T ELF-MF, which are the safety standards of public and occupational exposure for 2 h/day for 10 months. Brain tissues were immunohistochemically stained for the active (cleaved) caspase-3 in order to measure the apoptotic index by a semi-quantitative scoring system. In addition, the levels of catalase (CAT), malondialdehyde (MDA), myeloperoxidase (MPO), total antioxidative capacity (TAC), total oxidant status (TOS), and oxidative stress index (OS!) were measured in rat brain. Final score of apoptosis and MPO activity were not significantly different between the groups. CAT activity decreased in both exposure groups (p<0.05), while TAC was found to be lower in ELF 500 group than those in ELF-100 and sham groups (p<0.05). MDA, TOS, and OSI values were found to be higher in ELF-500 group than those in ELF-100 and sham groups (p<0.05). In conclusion, apoptosis was not changed by long-term ELF-MF exposure, while both 100 and 500 mu T ELF-MF exposure induced toxic effect in the rat brain by increasing oxidative stress and diminishing antioxidant defense system.Öğe IMPACTS OF SPECIFIC CRYOPROTECTANTS ON SPERM FREEZING AND RELATIONSHIPS BETWEEN CRYODAMAGE AND OXIDATION STRESS PARAMETERS IN AWASSI RAM SPERM(Cryo Letters, 2021) Varisli, Omer; Erat, Serkan; Bozkaya, Faruk; Aydilek, Nurettin; Taskin, AbdullahBACKGROUND: The role of oxidative stress during cryoprotectant treatment has received little attention. OBJECTIVE: To assess the effects of different cryoprotectants and discover relationships between cryodamage and oxidative stress parameters on Awassi ram sperm. MATERIALS AND METHODS: The sperm samples diluted with Salamon's tris-citrate (TRIS) containing 20% centrifuged egg yolk and 0.5, 1.0 or 1.5 M Glycerol (Gly), methanol (M), 2-methoxyethanol (2-ME), dimethylacetamide (DMA) and 1.2 propanediol (PR). After 2 h of equilibration at +4 degrees C, the sperm samples were frozen in liquid nitrogen vapour and stored. RESULTS: The best post-thaw motility (43.3%, 41.7%) of sperm was achieved when protected with 0.5 and 1.0 M glycerol. Arylesterase and ceruloplasmin parameters were significantly different after equilibration, whereas sulfhydryl groups were significantly different after freezing in their respective groups (P< 0.05). CONCLUSION: The increased use of glycerol caused greater loss of motility. The role of oxidative stress in freezing was also found to be limited.Öğe Oxidative parameters in bipolar patients treated with electroconvulsive therapy and pharmacotherapy(Yerkure Tanitim & Yayincilik Hizmetleri A S, 2013) Bulut, Mahmut; Altindag, Abdurrahman; Deveci, Zeliha; Kaya, M. Cemal; Bulbl, Feridun; Taskin, Abdullah; Kocamer, SengulElectroconvulsive (ECT) is one of the most efficacious choice in the treatment of bipolar disorder. There is no study conducted on human being investigating the effect of ECT on oxidative balance in bipolar disorder. In the present study, we aimed to evaluate the oxidative parameters on bipolar inpatients who is treated with ECT and pharmacotherapy. 31 bipolar inpatients in psychiatry department of Gaziantep University and 35 healty volunteeers were included in the study. Total Antioxidant Status (TAS), Total Oxidant Status (TOS) of serum samples were measured and Oxidative Stress Index (OSI) were counted in Harran University Biochemistry Laboratory. In all patient group, bipolar manic episode and depressive episode patients; before ECT and after ECT there was no significant difference for oxidative parameters. Before ECT, TAS levels in bipolar depressive episode patients were significantly lower than the control group (z=-2.614, p= 0.009). But after ECT, there was no difference between these two groups (z=-1.558, p> 0.05). Before ECT, TOS levels in bipolar manic episode patients were significantly higher than the control group (z=-2.598, p= 0.009); after ECT, there was no difference between these two groups (z=-0.910, p> 0.05). Finally, when compared with controls, in bipolar depressive episode patients TAS levels were increased with ECT and pharmacotheraphy and in bipolar manic episode patients TOS levels were decreased. For more comprehensive results new studies are needed.Öğe Paraoxonase Activity and Oxidative Status in Patients with Tinnitus(Korean Audiological Soc, 2016) Koc, Sema; Akyuz, Servet; Somuk, Battal Tahsin; Soyalic, Harun; Yilmaz, Beyhan; Taskin, Abdullah; Bilinc, HasanBackground and Objectives: The aim of this study was to investigate serum paraoxanase-1 (PON) activity, total oxidant status (TOS), total antioxidant status (TAS), and the oxidative stress index (OSI) in tinnitus; and to compare the results with data from healthy subjects. Subjects and Methods: A total of 114 subjects-54 patients with tinnitus and 60 healthy controls were enrolled in this study. Serum PON activity, TOS, TAS, and OSI levels were measured. Results: In the tinnitus group, TAS, and PON were significantly lower than in the control group (p<0.001). However, the TOS, and OSI levels were significantly higher in the tinnitus group than in the control group (p<0.001). Conclusions: According to the data obtained from the present study, patients with tinnitus were exposed to potent oxidative stress. Oxidative stress may be the key contributing factor to the pathogenesis of tinnitus.Öğe RELATIONSHIP BETWEEN TOXICITY OF CRYOPROTECTANTS, OSMOTIC AND OXIDATIVE STRESSES IN AWASSI RAM SPERM(Cryo Letters, 2022) Varisli, Omer; Bozkaya, Faruk; Aydilek, Nurettin; Taskin, AbdullahBACKGROUND: The relationship between the toxicity of cryoprotectants and their osmotic and/or oxidative stresses remains to be further investigated. OBJECTIVE: To investigate the toxic effects of different cryoprotectants and osmotic stress on Awassi ram sperm and to determine the relationship between oxidative and antioxidative status of the sperm. MATERIALS AND METHODS: Pooled sperm samples were exposed to sucrose solutions of different concentrations (75 to 900 mOsm) and isosmotic condition (290-325 mOsm) was re-established by adding HEPES buffered Tyrode's lactate. Sperm samples were mixed with 0.5, 1.0 and 1.5 M of glycerol, methanol, 2-methoxyethanol, dimethylacetamide or 1,2-propanediol for 5 min and returned to isosmotic condition. Sperm samples were exposed to cryoprotectants at 4 degrees C for 2 hours and isosmotic conditions were re-established. Motility, viability, acrosome integrity and oxidative or antioxidative parameters were determined. RESULTS: Treatment with hypo- or hyperosmotic sucrose solution reduced motility and viability without affecting acrosome integrity. The addition and removal of glycerol and dimethylacetamide (1.0 or 1.5 M) decreased sperm motility, while cryoprotectants had no effect on viability except for 1.5 M glycerol. Chilling significantly reduced the motility and viability of the sperm, but not the acrosome integrity. Rapid addition or removal of cryoprotectants also did not affect the acrosome integrity. Cryoprotectants changed only the ceruloplasmin level, while there were significant post-chilling differences in lipid hydroperoxide, paraoxonase and ceruloplasmin levels. CONCLUSION: Cryoprotectants without other additives have limited protection and glycerol can be toxic to spermatozoa. The oxidative stress plays a role in cryoprotectant toxicity and chilling stress.
