Öğe Effects of lycopene and vitamine E administration over gastric mucosal damage induced by Aflatoxin B1(2009) Kurt D.; Saruhan B.G.; Yokus B.; Cakir D.U.In the present study, we aimed to determine, the changes induced by Aflatoxin B1 (AFBj) administration on rat gastric mucosal barrier and gastric mucins alongside revealing the protective effects of lycopene and Vit E. Thirty-five Wistar-Albino male rats weighing 180-220 g, were divided into 7 groups as to include 5 rats in each group: control, lycopene (10 mg/kg/day lycopene for 15 days (Lycopene 10% FS), AFB, (single dose of 2.5 mg AFBj kg-1 on the 12th day of the study), Lycopene + AFB1 (10 mg lycopene/kg/day for 15 days and single dose of 2.5 mg AFB1 kg-1 on the 12th day of the study) and Vitamin E + AFB1, (10 mg/kg/day Vitamin E for 15 days and single dose 2.5 mg AFB1, kg-1 on 12th day). Following the sacrifice of study subjects on the 15th day, gastric mucus and phospholipid levels were determined and their stomachs were examined histopathologically. Examination of mucus and phospholipid levels revealed a significant reduction in group 3-5, in which AFB1 has been applied (respectively, p<0.001, p<0.001). When lycopene and vitamine E groups are compared with the AFB1 group, a significant elevation was detected in mucus and phospholipid levels (respectively, p<0.001, p<0.001). Whereas, histopafhological examination of gastric mucosas of the aflatoxin group showed degenerative changes, gastric mucosas of the control group and the remaining study groups were normal. Histochemically, while neutral mucins were predominant in general structure of stomach, mixed and sialomucins were observed, as well. Particularly acid mucins with suplhate and periodate reactive acid mucins were found to be more predominant in the aflatoxin group compared to control and other groups. Histochemical features of mucins were observed to be consistent with specific functions of the different regions of stomach. Lycopene and vitamine E administrations were found to be protective against the damage induced by aflatoxin on gastric mucosa. © Medwell Journals, 2009.Öğe Immunohistochemical localisation of oestrogen and epidermal growth factor receptors of the bitch uterus in the sexual cycle(2010) Ozyurtlu N.; Sagsoz H.; Saruhan B.G.; Zonturlu A.K.; Ketani M.A.; Akbalik M.E.The localisation of oestrogen (ER) and epidermal growth factor receptors (EGFR) in the various cell types of the bitch uterine was determined. In this study, 23 adult, healthy crossbred bitches brought to the clinic for ovariohisterectomy were used. ER and EGFR positive staining was detected in all cell types of the uterus. A distinct staining was seen in the luminal and glandular epithelium; while stromal and myometrial cells showed weak or moderate staining. The endothelial and smooth muscle cells of the vessels in the endometrium and myometrium sometimes appeared positive. No staining was observed in the mesothelium. The results of this study suggested that ER and EGFR were expressed at various levels in different cell types of bitch uterus. In light of the previous studies, and data of the presented investigations, it may be necesssary to elicit the harmonious proliferation and differentiation of epithelial and stromal cells that are considered essential for the preparation of the uterus for implantation.Öğe Radio-protective effects of melatonin on the skin histology in the rat(University of Dicle, 2014) Cakir Z.U.; Demirel C.; Ugurlu P.; Saruhan B.G.; Ketani S.; Seker U.; Zincircioglu S.B.In this study, the protective effect of melatonin on the radiation-damaged skin was investigated and effectiveness of the protective effect of melatonin was compared with amifostine. Fort this purpose, 40 female Sparague Dawley rats were used. Animals were divided into 5 groups. Control group (C), radiation group (R), and 2 experimental groups (radiation + melatonin 25 mg/kg (R+M) and radiation + amifostin 200 mg/kg (R+WR). A single dose of 20 gy gamma radiation were exposed to the left legs of the rat groups of R, R+M,and R+WR. At the end of a four-week of experiment period, the skin on the left femur of each rat was dissected and routine histological procedures were applied. Nikon Eclipse 400 research microscopy was used for histological observations. All histological structures of the skin were found to be normal in the control group. However, we observed that reduction in thickness of the epidermis and the dermal papillae have disappeared, sweat glands, sebaceous glands and hair follicles atrophy was observed in radiation groups. Our findings showed that R+M group, the thickness of the epidermis, collagen synthesis, R and R+WR group was better with histological structure. As a result, the histological abnormalities of radiation is shaped to protect in the skin, it has been found that melatonin has a significant effect compared to Amifostine (WR2721).
