Öğe Are resin-containing pulp capping materials as reliable as traditional ones in terms of local and systemic biological effects?(Japanese Soc Dental Materials Devices, 2022) Bakır, Elif Pınar; Yıldırım, Zehra Susgun; Bakır, Şeyhmus; Ketani, AydınThe aim of this study was to compare the local and systemic effects of current pulp capping materials containing resin with those of traditional materials in an animal study. A total of 48 rats were used: a control group (n=12) (sub-control and negative control), a resin-containing group (n=18) (Calcimol LC, Theracal LC, Activa-BioActive Base/Liner), and a traditional group (n=18) (Biodentine, ProRoot MTA, Dycal). The materials which had been placed in polyethylene tubes were implanted in subcutaneous pockets. The rats were sacrificed at 1, 2, or 4 weeks. Evaluations were made of subcutaneous connective tissue, the left kidney, liver, and blood samples. Of all the study groups, MTA demonstrated biocompatibility at a level close to that of the control groups. Inflammation was observed to be more severe in resin-containing materials, but Activa Base/Liner showed a more successful local and systemic tissue response.Öğe Can paricalcitol increase the effectiveness of N-acetylcysteine in contrast induced acute kidney prophylaxis in rats? A biochemical and histopathological study(Universidad de la Frontera, 2022) Yıldırım, Yaşar; Bahadır, Veysi; Aydın, Emre; Aydın, Fatma Yılmaz; Yılmaz, Zülfükar; Ketani, Aydın; Kaplan, İbrahim; Tuncer, Mehmet Cudi; Kadiroǧlu, Ali Kemal; Yılmaz, Mehmet EminN-Acetylcysteine (NAC) is used for contrast induced acut kidney injury (CI-AKI) prophylaxis because of its antioxidant effects. Paricalcitol, which has reno-protective effects, is likely to provide a more effective prophylaxis when added to NAC treatment. The study was designed based on this hypothesis. The study was organised to include 4 groups each consisting of 7 rats. Group 1 was the control group, and Group 2 included rats with CI-AKI. Rats in Group 3 were administered NAC at a dose of 100 mg/kg via oral gavage once a day for 5 days. Rats in group 4 were administered paricalcitol at a dose of 0.4 mcg/kg once a day for 5 days in addition to NAC. CI-AKI was induced after the treatments in both groups. The study was terminated on the sixth day. Samples were collected from the rats’ sera and kidney tissues to study oxidant and antioxidant parameters; kidney function tests were also studied. There were significant differences between the contrast nephropathy group (Group 2) and NAC and NAC+paricalcitol groups with respect to serum urea and creatinine levels. When the same groups were compared regarding oxidant (TOS-MDA) and antioxidant (TAC-Paraoxonase) parameters, we observed that the oxidant parameters increased in serum and kidney tissue samples with NAC use, and that effect was strengthened by the addition of paricalcitol to NAC treatment. However, despite increased antioxidant effectiveness, we observed no decrease in urea and creatinine levels when paricalcitol was added for CI-AKI in rats. There was no significant difference between Group 3 and Group 4. Paricalcitol provides a more potent antioxidant effect in both serum and kidney tissue samples when added to NAC treatment in rats with CI-AKI. Despite increased antioxidant parameters, however, paricalcitol does not provide a significant decrease in urea and creatinine levels.Öğe Effects of clinoptilolite on the digestibility of nutrients and relative organ weights in rat diets(Ankara Üniversitesi Ziraat Fakültesi, 2018) Demirel, Ramazan; Baran, Murat Sedat; Demirel, Dilek Şentürk; Ketani, Aydın; Yokuş, BeranThis study was conducted to compare the effects of dietary clinoptilolite on nutrient digestibility and relative organ weights in rats. In this study, 24 adult male Sprague-Dawley rats were divided into 4 groups with 3 replicates, according to a randomized split plots design. In addition to a control group (0% clinoptilolit), 3 levels of clinoptilolite (2%, 4%, and 6%) were used in the diets, and the rats were fed (individually in cages) these diets for 56 days. Statistically significant differences were found among the groups for the digestibility rates of nutrients (P<0.05), except for crude fibre (CF) and acid detergent fibre (ADF). The addition of clinoptilolite in rats’ feeds reduced the digestibility of crude fibre, crude ash (CA), neutral detergent fibre (NDF) and acid detergent fibre (ADF), but increased the digestibility of other nutrients. Slaughter live, organs (liver, kidney, heart, and stomach) and relative organ weights were not affected significantly by the clinoptilolite contents (P>0.05). In conclusion, clinoptilolites can be used in animal feed as natural toxin binders when stored under suitable conditions.Öğe Effects of fish oil hepatic peroxidation in rats and antioxidant role of vitamin E(2000) Canoruç, Fikri; Ketani, Aydın; Gürkan, Nihat; Mete, Nuriye; Canoruç, NaimeBu çalışmada balık yağı ile beslenen ratlarda hepatik lipid peroksidosyonunu nasıl etkilediği ve farklı dozlarda E vitamininin etkisi araştırıldı. Yöntem: 250±30 arasında değişen, 6 haftalık 32 erkek sprague-Dawley türü rat kullanıldı. Ratlar biri kontrol olmak üzere 4 gruba ayrıldı. Bütün gruplara diyetlerinin % 10'unu balık yağı oluşturacak şekilde bir diyet uygulandı. Ayrıca kontrol grubu dışındaki diğer üç gruba sırasıyla 3, 45, 209 IU/kg diyet olacak şekilde farklı dozlardaki E vitamini balık yağı ile birlikte verildi. 10 haftalık bir süreden sonra karaciğer dokusu alındı. Karaciğer dokusundaki lipid peroksidasyonu derecesi tiyobarbitürik asit yöntemi ile MDA tayini yapılarak bulundu. Ortalama değerler kontrol grubunda 148 mg/g doku, grup I'de 138, grup II'de 87.50 ve grup III'te ise 81.50 nmol/g doku olarak bulundu. Kontrol grubu ile karşılaştırıldığında grup II ve III'te lipid peroksidasyonu anlamlı bulundu (P<0.001). Grup I'deki yükselme, grup II ve III'e göre daha anlamlıydı (P<0.001). Grup II ve III arasında çok anlamlı bir fark görülmedi (P>0.005). Sonuç: Belli oranda balık içeren bir diyetle beslenen ratlar'da hepatik lipid peroksidasyonunun arttığı ve E-vitaminin bunda önemli bir koruyucu etkisinin olduğu görüldü. En etkin koruma 45 IU/kg diyet E-vitamini uygulanan grupta görüldü.Öğe Histologic effects of bilateral adrenalectomy under the light microscopy on rat mandibular condyle(2009) Ülkü, Sabiha Zelal; Zengingül, Ali İhsan; Ketani, AydınThe aim of this study was to compare the histologic findings from the condyles of rats with the general system findings. 30 Sprague-Dawley adult rats were used. The rats were sacrified and their condyles and femurs were examined histologically. The rats weight were observed weekly, their femoral height and weight, femoral mineral elements, and their daily food and water consumes were recorded. The results were analysed statistically by using SPSS (10.0). During the histologic examination of femur, the osteogenesis and calcification of the femur layers of the male/female rats in the experimental group were found to be lower than those of the ones in the control group. In addition to that, during the histological examination of condyle the level of thickness of condyle layers, the number of cellular elements, the osteogenesis and calcification seemed to be decreased for the male/female rats in the experimental group than for those in the control group. The decrease was found to be more significant for the female rats than the male rats in experimental group and it is thought that this is related to the gonad hormones. These findings emphasize that a reduction of sex hormone levels, induced by adrenalectomy, significantly influences condylar histogenesis.Öğe Identification of Vibrio parahaemolyticus by PCR Method in Fish and WaterSamples from Tigris River, Turkey(2021) Ketani, Aydın; Özcan, Filiz; Arserim, Neval BerrinClimate change has caused increasing Vibrio parahaemolyticus infections in areas unaffected by this pathogen.Water temperature is an important ecological factor. It is consequently of high importance to follow V. parahaemolyticus inaquatic environments and foods. The aim of this work was to detect the presence of V. parahaemolyticus in Tigris Riverfrom eight different sites namely Malabadi Bridge, Fiskaya, Ongözlü Bridge, Bağıvar, Pamukçay, Ambarçay, Seyrantepe andSilvan. Total 480 samples of water and fish were used for V. parahaemolyticus detection. Thiosulphatecitrate?bile saltsucrose (TCBS) Agar was employed for bacteria isolation for 24 h at 37?C. The dominant uniform bacterial colonies werepurified by streaking onto the TCBS agar plates three times. Identification of this pathogen was made by PCR. A total of 480water and fish samples were analyzed. According to the results, V. parahaemolyticus was detected in 40 (6.6%) fish samplestaken from the Seyrantepe region.Öğe The protective effects of vitamin C, vitamin E and vitamin E+ glutathion on the sperm number, sperm form and testes of irradiated rats(2000) Akdağ, Zülküf; Deniz, Mustafa; Ketani, Aydın; Çelik, Mustafa Salih; Sert, CemilBu çalışmada, X-lşınlarının neden olduğu hasarlara karşı, Vitamin C, Vitamin E ve Vitamin E+Glutathion karışımının koruyucu etkileri araştırıldı. 200-220 gr ağırlığında, toplam 50 Wistar Albino rat beş gruba ayrılarak kullanıldı. Lgrup sham grubu, 2. grup yal¬nızca ışınlanan grup, 3,4,5. gruplar radioprotektif ajan verilen ve sonradan ışınlanan deney gruplarıydı. Rafları radyasyona maruz bırakmazdan önce 3. gruba Vit C(400mg/kg/gün), 4.gruba Vit E (400 mg/kg/gün), 5.gruba Vit E+glutathion (400+400 mg/kg/gün) 15 gün süreyle verildi. Tüm deney guru¬pları ve kontrol gurubu 8.9 Gy X-lşınına maruz bırakıldı. Radyasyona maruz bırakıldıktan bir gün sonra tüm raflar sakrifiye edildi. Sertoli ve Leydig hücreleri, spermatogonia hücre histopatolojisi ve sperm hücrelerinin anormalliği kontrol edildi. Yalnızca ışınlanmış grupta, Leydig ve spermatogonia hücre sayısında azalma, Sertoli hücre sayısında artış gözlendi. Deney gruplarında Sertoli hücre sayısındaki artış devam ederken, Leydig ve spermatogonia hücre sayısında azalma gözlendi. Kontrol grubuna kıyasla, yalnızca ışınlanmış grupta sperm sayısı ve anormalliğinde belirgin bir değişim gözlendi. Ancak, bu değişimler yalnızca ışınlanmış gruba kıyasla, Vitamin C, Vitamin E ve Vitamin E+Glutathion uygulanan gruplarda gözlenmedi.Öğe Rats with Acute Gastritis Experimentally Induced with Ethanol Distribution of Mast Cells in the Liver, Lung and Kidneys(Dokuz Eylul University, 2024) Aydın, Muhammed Kasım; Baksi, Tuba; Karakoç, Zelal; Baksi, Nazan; Ketani, AydınMast cells are found in systems that are in contact with the external environment, such as blood vessels, nerves, respiration, and digestion. Mast cell proteases play important roles in the regulation of humoral and cellular events in the tissue. In rodents, mast cells are of two types: mucosal and connective tissue origin. This study aimed to examine mast cells histochemically and eosinophils in the possible damage that ethanol may cause in the liver, lung, and kidneys of rats with gastritis and also to determine whether pomegranate extract affects these cells. In our study, liver, lung, and kidney tissue samples taken from rats were stained with Alcian Blue/ Safranin O, Toluidine Blue to identify mast cells, and Congo Red to identify eosinophils. In the histological evaluation, it was determined that mast cells were located especially around blood vessels. It was determined that heparin-containing mast cells were dense in liver tissue, and histamine-containing mast cells were dense in lung tissue. It was determined that mast cells were less dense in kidney tissue than in liver and lung tissues. When the locations and densities of mast cells in rats are considered, it can be said that they undertake similar tasks in functions such as vasopermeability and inflammatory cell response, as in other mammalian species.Öğe Veteriner fakültesi öğrencilerinin mikroskop kullanımına yönelik öz yeterlik inançlarının belirlenmesi(Dicle Üniversitesi Ziya Gökalp Eğitim Fakültesi, 2020) Sağsöz, Hakan; Yapıcı, İ. Çalışmada tarama modeli kullanılmıştır. Çalışma grubunu; 2018-2019 öğretim yılında Dicle Üniversitesi Veteriner Fakültesi’nde öğrenim gören 77’si kadın, 119’u erkek olmak üzere toplam 196 öğrenci oluşturmaktadır. Veri toplama aracı olarak Ünal Baş (2013) tarafından geliştirilen “Mikroskop Kullanımında Öz yeterlik İnanç Ölçeği” kullanılmıştır. Verilerin analizinde; %, frekans, ortalama, standart sapma, bağımsız gruplar t-testi ve tek yönlü varyans analizi (ANOVA) kullanılmıştır. Çalışma sonucunda; öğrencilerin ölçeğin geneline ilişkin aldıkları toplam puan ortalamalarına göre (116,72) öz yeterlik inanç düzeylerinin “yüksek” olduğu belirlenmiştir.
