Öğe Brucellosis Seroprevalance on Goats, Sheep, Cows and Man who are under Risk in Diyarbakir Region(Medwell Online, 2012) Arserim, N. B.; Ozekinci, T.; Ceylan, A.; Tel, O. Y.; Keskin, O.; Vural, A.The aim of this study was to determine the prevalence of brucellosis in sheep, goats, cows and human who are at risk in Diyarbakir region. From ten locations obtained in Diyarbakir, blood samples were collected from sheep, goats and cows belonging to different herds and man who are at risk between 2007 and 2009 years. The blood samples were tested using the Microagglutination (MAT) technique. Seropositivity by using MAT technique on the collected blood samples were positive on 60 (35.2%) of 170 sera taken from the veterinarian, slaughterhouse staff, butchers and growers. Also the positivity rates were 20 (2.9%) of 680, 5 (0.7%) of 725 and 25 (4.3%) of 575 for sera samples taken from cows, sheep and goats, respectively. Consequently, seropositivity is obtained high for animals and individuals who are under risk according to the other regions in Turkey. E.; Soyer, O. U.; Keskin, O.; Uzuner, N.; Yazicioglu, M.; Kilic, M.; Artac, H.Introduction The reliability and validity of Turkish version of Childhood Asthma Control Test (C-ACT). Purpose The management of asthma is an important as well as difficult issue of physician's daily practice particularly in busy clinical settings. C-ACT was created to identify asthma control levels in children aged 4-11 years. Our aim was to evaluate the reliability, validity and responsiveness of C-ACT in a Turkish sample of children with asthma. Method In this multicenter study, 368 children were enrolled. C-ACT was completed every month by parents and patients who were evaluated in 3 visits within 2 month intervals. At each visit, physicians interpret the control level and decided for the treatment step as established in GINA guidelines. Results The internal consistency reliability of the Turkish version of C-ACT (C-ACT1 to C-ACT5) was found to be 0.82, 0.83, 0.82, 0.82 and 0.80, respectively (reliability statistics, Cronbach's alpha). Test-retest reliability was 0.71. There was significant correlation between C-ACT and physician's assessment of asthma control at visit 1 (r = 0.65, P < 0.001). Conclusions Turkish version of C-ACT is an accurate and reliable tool to evaluate asthma control in children aged 4-11 years. Its widespread use may facilitate appropriate assessment of asthma control and may lead to decrease the number of uncontrolled patients.Öğe Seroepidemiological survey of Rhodococcus equi infection in asymptomatic horses and donkeys from southeast Turkey(South African Vet Assoc, 2011) Tel, O. B.; Keskin, O.In order to assess the level of Rhodococcus equi infection in southeast Turkey, 679 sera from healthy foals and adult horses and 78 sera from donkeys were tested by indirect ELISA using a R. equi reference strain (ATCC 33701) as antigen. Eighty (11.7 %) sera from horses and 9(11.5 %) sera from donkeys with titres >0.85 were positive. The prevalence of seropositive horses in Sanliurfa Province was higher than in Diyarbakir Province; 56(13.9 %) horses in Sanliurfa Province and 24(8.7 %) horses in Diyarbakir Province were defined as seropositive. In Sanliurfa Province 14.5 % of female (n = 343) and 10.1 % of male (n = 59) horses tested were defined as seropositive, while in Diyarbakir Province more males (11.4 %, n = 114) were seropositive than females (6.7 %, n = 163). Horses 1 to 5 years of age were found to have the highest seropositivity rate in both provinces. A total of 78 sera from donkeys were investigated in Sanliurfa Province, of which 9 (11.5 %) were positive by ELISA. Among the 9 positive sera, 6(12.8 %) were from donkeys 1-5 years old and 3(13.6 %) were from donkeys >5 years of age. No positive sera were found in donkeys less than 1 year old. Five(12.5 %) sera of females and 4(10.5 %) sera of males tested were positive. These results indicate the existence of R. qui in the horse populations in Sanliurfa and Diyarbakir Provinces. Similar infection rates were found for donkeys in Sanliurfa. This suggests the importance of serological surveys to diagnose R. equi infection in the region and to prevent the zoonotic risk.Öğe Tumour necrosis factor-alpha, interleukin-10, interferon-gamma and vitamin D receptor gene polymorphisms in patients with chronic hepatitis delta(Wiley, 2014) Karatayli, S. A.; Keskin, O.; Karatayli, E.; Albayrak, R.; Ozkan, M.No data exist to assess certain polymorphisms that have a potential effect on the immune response in patients with chronic hepatitis delta (CHD). The aim of this study was to investigate polymorphisms in 6 polymorphic sites: IL-10 -1082 (rs1800896), IL-10 -627 (rs1800872), IFN-gamma +874 (rs62559044), TNF-alpha -308 (rs1800629), vitamin D receptor (VDR) FokI (rs2228570) and VDR TaqI (rs731236). The genotypes of 67 patients with CHD and 119 patients with chronic hepatitis B (CHB) were compared. In addition, 56 individuals with resolved hepatitis B virus (HBV) infection were used as a control group for patients with CHB. Polymorphisms in TNF-alpha, IL-10, and VDR genes were analysed using polymerase chain reaction/restriction fragment length polymorphism methods. The IFN-gamma gene polymorphism was detected by allele-specific polymerase chain reaction (PCR). Patients with CDH were more likely to have advanced liver disease compared with patients with CHB (P<0.0001). IL-10 -1082 and VDR TaqI polymorphisms showed significant differences between patients with CHD and CHB. The high secretory IL-10 -1082 genotype GG was less frequent in CHD compared with patients with CHB and resolved HBV (17.7%, 37.4% and 47.1%, respectively (PCHD vs CHB and resolved HBV). The frequency of the high secretory VDR TaqI TT genotype was 86.6% in patients with CHD, 62.7% in patients with CHB and 62.5% in resolved HBV individuals (CHD vs CHB: P<0.05). None of the polymorphisms analysed had an effect on HBV persistence.
