Öğe 68Ga-PSMA Versus 18F-FDG PET/CT in the Evaluation of Epithelioid Hemangioendothelioma(Lippincott Williams & Wilkins, 2024) Senses, Veysi; Kaplan, Ihsan; Kaya, Fulya; Ibiloglu, Ibrahim; Can, CananA 32-year-old man with epithelioid hemangioendothelioma was imaged with Ga-68-PSMA PET/CT to evaluate the potential of Lu-177-PSMA radionuclide therapy after progression findings were observed on follow-up F-18-FDG PET/CT imaging. Liver lesions showed low FDG SUVmax. The lesion-to-background ratio (TBR) was calculated as 5.1/2.9 on F-18-FDG PET/CT imaging. However, Ga-68-PSMA PET/CT showed moderate PSMA SUVmax. TBR was calculated as 4.5/1.1. In conclusion, in this case report, we demonstrated that Ga-68-PSMA with moderate TBR in epithelioid hemangioendothelioma may be promising for Lu-177-PSMA radionuclide therapy.Öğe Anti-inflammatory and anti-oxidant role of ursolic acid in cerulein-induced acute pancreatitis in rats(Acad Nacional Medicina Mexico, 2023) Turkoglu, Ahmet; Ibiloglu, Ibrahim; Kaplan, Ibrahim; Arslan, Serkan; Halil-Ocal, Ibrahim; Gumus, MetehanBackground: Ursolic acid (UA) is found in many plants, and has been reported to have anti-protease, antioxidant, anti-inflammatory, antimicrobial, nephroprotective, hepatoprotective, and cardioprotective effects. Objective: The purpose of this study was to investigate the effects of ursolic acid in cerulein-induced acute pancreatitis (AP). Materials and methods: Thirty-two Wistar albino rats were randomly assigned to 4 equal groups: Sham, acute pancreatitis, treatment, and ursolic acid group. Results: Serum amylase levels in the AP and treatment groups were significantly higher than in the others (p < 0.05). In addition, serum IL-113, IL-6, and TNF-& alpha; levels were significantly higher in the AP group in comparison with the treatment group. Although pancreatic tissue total oxidant activity in the AP and treatment groups was similar, pancreatic tissue total antioxidant capacity was significantly higher in the treatment group than in the AP group. Conclusions: Damage to the pancreas and remote organs in AP was observed to be reduced by UA. This disease is, unfortunately, at its advanced stage when it is diagnosed. Survival time is usually not more than a few months. The aim of this study was to analyse the expression of Aquaporin 1, Aquaporin 3 and Aquaporin 5 in malignant pleural mesotheliomas and to explore the relationship of these levels of expression with epidermal growth factor receptor (EGFR) gene mutation and prognostic parameters. In this study, 60 cases diagnosed as malignant pleural mesothelioma among the pleural biopsy materials in the archives of the Pathology Department of Medical Faculty of Dicle University in 2003-2013 were evaluated. The tissues were stained immunohistochemically with antibodies against Aquaporin 1, Aquaporin 3 and Aquaporin 5, and the existence of EGFR mutation was investigated in the tissues by real-time polymerase chain reaction (PCR). The obtained results showed expression of Aquaporin 1, Aquaporin 3 and Aquaporin 5 in varied amounts in malignant pleural mesotheliomas. However, no significant relation was obtained thus far between the expression levels of these aquaporins and the prognostic parameters. No mutations were detected in the EGFR gene exons 18-21 by using real-time PCR. It could be suggested that although Aquaporin 1, Aquaporin 3 and Aquaporin 5 are expressed in malignant pleural mesothelioma, they do not have any effect on the prognostic parameters. Mutations in different domains of EGFR gene, other than exons 18-21, should be sought to develop new targeted treatments.Öğe Can aquaporins be used as diagnostic and prognostic markers for uterine smooth muscle tumours?(Taylor & Francis Ltd, 2017) Alabalik, Ulas; Turkcu, Gul; Keles, Ayse Nur; Ibiloglu, Ibrahim; Ozler, Ali; Urakci, Zuhat; Buyukbayram, HuseyinGenerally, uterine leiomyosarcoma is easily diagnosed. However, uterine smooth muscle tumours which show atypical histological features and unusual growth patterns may mimic malignancy and may not be easily diagnosed. In this study, our aim is to show the expressions of Aquaporin3, Aquaporin7 and Aquaporin9 in uterine smooth muscle tumours, and to investigate if aquaglyceroporins can be used as diagnostic and prognostic markers to start rapidly an appropriate treatment for patients with these tumours in order to extend the survival time. We determined that there had been 74 patients diagnosed with uterine smooth muscle tumours. We divided patients into four groups based on the diagnosis: bizarre leiomyoma, smooth muscle tumour of uncertain malignant potential, leiomyosarcoma and leiomyoma. Aquaporin3, Aquaporin7 and Aquaporin9 were detected by using monoclonal anti-Aquaporin3, anti-Aquaporin7 and anti-Aquaporin9 antibodies, respectively. In leiomyosarcoma group, we observed a statistically significant relation of Aquaporin3 expression with survival time, grade, stage, mitotic index and Ki-67 score. A significant relation of both Aquaporin7 and Aquaporin9 expressions with survival time, grade, stage was not statistically detected in leiomyosarcoma group. The decrease of Aquaporin3 expression can be used as important diagnostic and prognostic marker. Aquaporin7 and Aquaporin9 expressions cannot be used as diagnostic and prognostic markers.Öğe Colchicine may be of therapeutic benefit in prurigo pigmentosa(Wiley, 2018) An, Isa; Ucmak, Derya; Ibiloglu, Ibrahim; Demir, Vasfiye; Akdeniz, SedatPrurigo pigmentosa (PP) is a rare inflammatory skin disease. Neutrophil-mediated inflammation is considered to be responsible for the etiopathogenesis of PP. We consider that colchicine may be an effective agent in the treatment of PP since it exerts an antiinflammatory effect by inhibiting neutrophil chemotaxis. Further studies are required to verify whether colchicine is an effective treatment option for PP.Öğe COMPARISON OF SKIP EXPRESSION IN MALIGNANT PLEURAL MESOTHELIOMAS WITH KI-67 PROLIFERATION INDEX AND PROGNOSTIC PARAMETERS(Vesalius Univ Medical Publ, 2016) Turkcu, Gul; Alabalik, Ulas; Keles, Ayse Nur; Ibiloglu, Ibrahim; Kucukoner, Mehmet; Sen, Hatice Selimoglu; Buyukbayram, HuseyinWe aimed to determine the presence of SKI-interacting protein (SKIP) expression in malignant pleural mesothelioma (MPM) and its effect on prognosis by investigating SKIP correlation with the Ki-67 proliferation index and prognostic parameters. Pathological preparations of the patients diagnosed with MPM between 2006 and 2012 were evaluated. Immunohistochemical analyses were performed to evaluate the expression of SKIP and the Ki-67 proliferation index. Correlations between SKIP expression, clinicopathological factors and survival were investigated. Survival data were calculated using the Kaplan-Meier method, and Cox regression analysis was used to evaluate the prognostic value of the variables. In total, 52 patients were evaluated in the study; 36 of them were male and 16 were female. The mean age of the patients was 62.3 +/- 12.2 years. The median overall survival period was 8.5 months. Factors negatively affecting general survival in the univariate analysis included high SKIP expression, Ki-67 proliferative index over 30%, presence of non-epithelioid type MPM and stage III-IV disease (p < 0.05). Cox regression analysis revealed that high SKIP expression, high Ki-67 proliferative index and presence of non-epithelioid type MPM are independent factors that affect the survival rate. Higher SKIP expression is associated with poor prognosis in MPM.Öğe Cordycepin prevents postoperative formation of intra-abdominal adhesion in a rat model: An experimental study(Turkish Assoc Trauma Emergency Surgery, 2017) Arslan, Serkan; Zeytun, Hikmet; Basuguy, Erol; Ibiloglu, Ibrahim; Uygun, Ibrahim; Yilmaz, Ahmet; Tan, IlhanBACKGROUND: The aim of the present study was to investigate whether cordycepin prevented adhesion formation in a rat model. METHODS: Rats were randomly assigned to 3 groups of 10 rats. Control group: The absence of adhesion was confirmed via laparotomy. Adhesion group: The cecum was removed from the abdomen and scraped with a dry gauze bandage until petechial hemorrhagic foci developed. Cordycepin group: The same surgical procedure was performed as in the adhesion group, and 10 mg/kg cordycepin was administered intraperitoneally. After 15 days, the rats were sacrificed humanely via cardiac blood withdrawal under anesthesia. The rats were then analyzed morphologically and histopathologically, and hydroxyproline (OH-p) and malondialdehyde (MDA) levels were measured. RESULTS: Macroscopic analysis revealed significantly less adhesion in the cordycepin group than in the adhesion group (p<0.01). Furthermore, significant histopathological improvement was also evident in the cordycepin group compared to the adhesion group (p<0.05). The levels of OH-p and MDA in blood and tissue were higher in the adhesion group than in the control group, and lower in the cordycepin group than the adhesion group. Interestingly, MDA level was significantly lower (blood: p<0.05; tissue: p<0.01) in the cordycepin group than in the adhesion group, whereas only tissue OH-p was significantly lower in the cordycepin group compared with the adhesion group (p<0.05). One rat in both adhesion group and cordycepin group died postoperatively. CONCLUSION: Results indicated that cordycepin effectively reduced adhesion in a rat abrasion model. In our study, we analyzed the histopathological and biochemical changes of low-dose Cordycepin(c) on a rat model in the kidney. Materials and methods: Twenty-four male Wistar Albino rats were randomly allocated to three groups (n = 8): the sham-control group (Group 1), the renal I/R-untreated (Group 2) group, and the I/R-C-treated (Group 3) group. Cordyceps was administered intraperitoneally at 5 mg/kg twice. Renal histological changes were compared and the relevant parameters of oxidative stress and inflammation were detected. Results: In blood and tissue biochemistry, it was observed that IL-1 Beta, IL 6, TNF alpha, MDA, TOS, and OSI increased in Group 2 and decreased in Group 3. It was determined that TAS values were increased in Group 3, and decreased in Group 2. In the histopathological evaluation, while Group 1 was evaluated as normal, significant kidney damage was detected in Group 2. It was determined that there was a significant decrease in kidney damage in Group 3. Conclusion: These results suggest that low dose Cordycepin was as effective as normal dose on renal ischemic reperfusion and reduction of damage.Öğe Effects of Ecballium Elaterium on Proinflammatory Cytokines in a Rat Model of Sepsis(Taylor & Francis Inc, 2016) Arslan, Mehmet Serif; Basuguy, Erol; Ibiloglu, Ibrahim; Bozdemir, Eda; Zeytun, Hikmet; Sahin, Atalay; Kaplan, IbrahimObjective: Ecballium elaterium (EE) is a plant from Cucurbitaceae family. Its anti-inflammatory role in sepsis is not well understood. We investigated the effects of EE on serum levels of proinflammatory cytokines and further explored the mechanisms underlying histological changes in liver and ileum following EE administration in a polymicrobial sepsis model. Methods: Thirty rats were divided into three groups of 10 rats each. Rats were subjected to sham laparotomy plus normal saline administration (control group, CG), laparotomy with cecal ligation and puncture (CLP) (sepsis group, SG), and laparotomy with CLP plus 2.5mg/kg EE administration (experimental group, EG). Twenty-four hours after laparotomy, animals underwent cardiac puncture, and blood was collected for interleukin 1 (IL-1), interleukin 6 (IL-6), and tumor necrosis factor alpha (TNF-) assessment. Whole sections of liver and ileum tissues were collected for histologic examination. Results: The serum level of IL-6 was significantly lower in EG as compared to SG. Although IL-6 levels were shown a statistically significant (p < 0.0001) decline to near control values, no significant changes were observed in serum levels of IL-1 and TNF- after EE treatment. Histologic examination revealed statistically significant reduction in collagen formation (p = 0.001) on serosal surface of ileum and hepatic venous congestion (p = 0.040) in EG as compared to SG. Roflumilast has been shown to be a potent anti-inflammatory and antioxidant agent. In the present study, we aimed to investigate the effect of roflumilast in cerulein-induced AP. Thirty-two male rats were divided into four groups: group 1 (sham), group 2 (Roflumilast), group 3 (AP), and group 4 (AP + Roflumilast). AP was induced by injecting 4 x 75 mu g/kg of body weight at an interval of 1 h. Rats were killed after 12 h following the last cerulein administration. AP was confirmed by measuring the serum amylase level and inflammatory features. Morphological changes were observed in the pancreas. Amylase levels were higher in the AP and AP + Roflumilast groups than the sham and Roflumilast groups. The serum levels of TNF-alpha, IL-1 beta, and IL-6 increased in the AP group, whereas they decreased in the Roflumilast group. The total oxidant activity (TOA) was higher and the total antioxidant capacity (TAC) was lower in the AP group. The administration of roflumilast decreased the TOA and increased the TAC in comparison with the AP group (p < 0.05 for both). Materials and methods: We retrospectively examined the pathology reports of 1841 patients who underwent surgeries in the general surgery clinic for the treatment of gallbladder diseases between March 2006 and May 2015 and whose gallbladder materials were examined after surgery. Age, gender, pathological diagnosis and presence of gallbladder stones were recorded. Results: The average age of the 1841 patients (male/female: 585/1256) evaluated in this study was 50.37 +/- 18.81 years (range: 2-97 years). Surgery for gallbladder disease was most commonly performed in the age range of 50-60 years. Chronic cholecystitis was the most common histopathological diagnosis (92.0%), followed by acute cholecystitis (5.1%), non-neoplastic lesions (1.74%), preneoplastic lesions (0.76%), carcinomas (0.33%) and lymphomas (0.1%). One or more gallbladder stones were detected in 955 patients. Of these, 93.4% and 4.1% patients were diagnosed with chronic cholecystitis and acute cholecystitis, respectively. Conclusion: Histopathological examination of gallbladder materials is important for identifying incidental benign and malignant tumours.Öğe Is There a Difference Between the Direct Immunofluorescence Slides Stored at Room Temperature in Bright Versus Dark Conditions?(Sci Printers & Publ Inc, 2021) Ibiloglu, IbrahimOBJECTIVE: Direct immunofluorescence (DIF)-stained slides are archived in the dark to prevent fading. Reevaluation of slides for clinical evaluation and training may be needed. In this study we aimed to determine the duration of archiving in dark environment (DE) versus bright environment (BE) in order to evaluate DIF positive stained slides. STUDY DESIGN: A total of 56 immunofluorescence slides consisting of 35 kidney and 21 skin biopsies were included in the study. RESULTS: The mean wilting time of the slides was 7.5 weeks for those left in the BE and 7.1 weeks for those in the DE. The mean wilting times of the kidney slides left in BE and DE were 6.8 and 7.2 weeks, respectively. The mean wilting times of the skin slides left in a BE and DE were 8.2 and 7 weeks, respectively. CONCLUSION: We did not observe a significant difference between immunofluorescence (IF) slides archived in light versus dark media. METHODS: A total of 308 patients diagnosed with lung cancer were included in this study. These 308 patients were first classified as group 1 (SPN 30 mm>longest lesion diameter >= 10 mm) and group 2 (lung mass (longest lesion diameter >= 30 mm), and then the same patients were classified as group 3 (nodular diameter of <= 20 mm) and group 4 (nodular size of >20 mm). Group 1 was compared with group 2 in terms of extrathoracic metastases. Similarly, group 3 was compared with group 4 in terms of frequency of extrathoracic metastases. F18 fluorodeoxyglucose positron emission tomography/CT examination was used to detect liver, adrenal, bone, and supraclavicular lymph node metastasis, besides extrathoracic metastasis. RESULTS: Liver, bone, and extrathoracic metastasis in group 1 was statistically lower than in group 2 (p<0.001, p<0.01, and p=0.03, respectively). Liver, extrathoracic, adrenal, and bone metastasis in group 3 was statistically lower than that in group 4 (p<0.001, p=0.01, and p=0.04, p<0.01, respectively). The extrathoracic extension was observed in only one patient in group 3. In addition, liver, adrenal, and bone metastases were not observed in group 3 patients. CONCLUSION: Positron emission tomography/CT may be more appropriate for cases with a nodule diameter of <= 20 mm. Performing local imaging in patients with a nodule diameter of <= 20 mm could reduce radiation exposure and save radiopharmaceuticals used in positron emission tomography/CT imaging.Öğe A new experimental rat model of pancreatitis using Ecballium elaterium(Elsevier, 2015) Arslan, Serkan; Okur, Mehmet Hanifi; Zeytun, Hikmet; Basuguy, Erol; Ibiloglu, Ibrahim; Turkoglu, Ahmet; Bozdag, ZubeyrBackground: We originally aimed to determine the beneficial effects of Ecballium Elaterium (EE) on acute pancreatitis; however, we observed negative effects of EE on the pancreas. Thus, we used EE in rats to generate a new model of pancreatitis, which we compared with other established pancreatitis models. Methods: A total of 32 Wistar albino rats were used. Rats were divided into 4 groups, each of which contained 8 rats. Group 1 (Control), Group 2 (L-Arginine (LA), Group 3 (LA + EE), Group 4 (EE): Twenty-four hours after that serum parameters were analyzed in the collected blood. Blood samples were transported on mice to the Biochemistry Laboratory following cardiac puncture. The levels of amylase, interleukin (IL)-6, interleukin (IL) 1-beta (IL-1 beta), malondialdhyde (MDA), tumor necrosis factor (TNF)-alpha, total antioxidant status (TAS), and total oxidant status (TOS) were analyzed. Histopathological analysis: The pancreas and lung tissue samples obtained from the rats. Edema, inflammation, vacuolization, and necrosis of the pancreas were assessed using a scoring system ranging from 0 to 4. Edema, hemorrhage and inflammation of the lung tissue were evaluated using a scoring system ranging from 0 to 3. Results: Histopathological analysis revealed that edema, inflammation, necrosis, and hemorrhage were significantly higher in the LA + EE group than in the control group. Moreover, necrosis was higher in the rats that received LA and EE compared to the rats that received only LA or EE. Increases in inflammatory mediator levels, including IL-6, IL-1 beta, TNF-alpha, MDA, and TOS, were observed in all groups as compared to the control group. Moreover, lower TAS levels were detected in all groups but the control group. The increase in IL-1 beta and TNF-alpha levels and the decrease in TAS were statistically significant in all groups (P < 0.05). Conclusions: EE may be used to create a successful acute pancreatitis (AP) model, resulting in edema, necrosis, hemorrhage, and inflammation of the pancreas. The major advantage of this model is that it does not require laparotomy, and can be implemented with only an intraperitoneal injection (IP). Moreover, EE may be combined with other agents, such as LA, to create severe pancreatitis. Further molecular studies are warranted to determine the underlying effects of EE on the pancreas. There are three histologic subtypes of mesothelioma: epithelioid, biphasic, and sarcomatoid. Pleural mesothelioma is usually characterized by diffuse pleural thickening. Disease progression is characterized by local invasion of the chest wall and lung. Lymphatic metastasis is rare and hematogenous metastasis is much rarer. The purpose of these case reports is to emphasize that pleural mesothelioma metastases can occur in unexpected places and to contribute to the literature.
