Öğe BIOCHEMICAL, HISTOPATHOLOGICAL AND IMMUNOHISTOCHEMICAL EVALUATION OF ALTERATIONS IN BLOOD AND TIBIAL BONE TISSUES OF OVARIECTOMIZED RATS IN THE ACUTE PHASE(Afyonkarahisar Sağlık Bilimleri Üniversitesi, 2020) Baloğlu, Murat; Gökalp-özkorkmaz, EbruOBJECTIVE: Postmenopausal osteoporosis results in decreased bone mass and deterioration of bone tissue. symptoms of osteopo-rosis are observed after ovarectomy and due to the fact that bone resorption is more than bone formation, it causes bone loss. ADAMs transmembrane proteins are involved in many physiological proces-ses, including reconstruction of the extracellular matrix and embr-yonic development, cell migration, adhesion, and angiogenesis. The aim of our study was to investigate the biochemical, histopathologi-cal and immunohistochemical alterations in blood values and tibia bone tissue of ovariectomized rats in the acute phase.MATERIAL AND METHODS: Twenty healthy and female adult Wis-tar albino rats were divided into two groups as control (n:10) and bilateral ovariectomy (n: 10). A 2-cm incision was performed from the ovarian cervix under anesthesia, and two dorsolateral incisions were made from the uterine apex and the bilateral ovaries were re-moved with a silk thread. One week after ovariectomy, experimental animals were sacrificed. Blood samples were taken from each ani-mal to determine serum estrogen, calcium and alkaline phosphate values. Tibia bones were removed and placed into formalin solution for fixation, then they were decalcified and paraffin blocks were pre-pared after routine histological procedures. Semi-thin sections were cut and stained with Hematoxylin-Eosin for histopathological exa-mination. For immunohistochemical analysis, the antigen retrieval process was performed with hydrogen peroxide after administrati-on to citrate buffer. Anti-ADAMTS-1 primary antibody was applied to the sections and evaluated under a light microscope.RESULTS: In the control group, while the Haversian and Volkmann canals were distributed in raider style, collagen fibers were in pa-rallel and regular. In the ovariectomy group, degenerative changes in the bone matrix, deterioration of lamellar bone structure were observed. ADAMTS-1 expression was positive in the control group osteoblast and osteocyte cells and fine collagen fibers in the bone matrix. In the ovariectomy group, increased ADAMTS-1 expression in osteoclast cells in the degenerative area, ADAMTS-1 positive exp-ression in endothelial cells was observed in osteon structures. There was a significant decrease in ovarian estrogen values in the ovarie-ctomy group compared to the control group. Calcium values of the control and ovariectomy groups were close to each other. Alkaline phosphatase levels were increased in the ovariectomy group (P = 0.01).CONCLUSIONS: In the light of data we obtained from our study, we suggest that the inclusion of ADAMTS-1 protein may support the new treatment methods in the treatment of osteoporosis.
