Öğe The effect of saline and different growth conditions on seed germination of almond grown in vitro and in vivo(International Society for Horticultural Science, 2014) Işıkalan, Çiǧdem; Akbaş, Filiz; Karakuş, Pınar; Kuru, İbrahim SelçukThe study aims to evaluate the role of germination conditions for almond [Prunus dulcis (Mill.) D.A. Webb] seeds. For the experiments in vitro germination, sterile almond seeds were cultured on 1/4 MS+100 mM NaCl, 1/4 MS (Murashige and Skoog) medium and for the control group on agar medium (agar and water only). The media used in the experiments was supplemented with 30 g/L sucrose and with 7 g/L agar. Data on the shoot number and the length of seedlings, the root number and the length were recorded after three weeks. The results indicated that the highest germination percentage was achieved on the agar-water medium (100%) or on 1/4 MS medium (92%). It was also found that the seed germination percentage was negatively affected by salinity (100 mM NaCl). A continious yellowing and necrosis on germinated seedling were observed too. The same conditions (1/4 MS+100 mM NaCl, 1/4 MS and tap water) was applied to the experiments in vivo. For this purpose seeds were planted on a mixture of peat and sand (2:1 w/w). The germination percentage of the seeds was found to be very slow and weak in all tested groups.Öğe Physiological and biochemical responses of rice (Oryza Sativa L.) varieties against drought stress(Bangladesh Botanical Society, 2021) Kuru, İbrahim Selçuk; Işıkalan, Çiğdem; Akbaş, FilizPhysiological and biochemical responses of six different rice (Oryza sativa L.) varieties cultivated in Turkey and two local varieties, namely Karacadag and Hazro to the drought were investigated. After 12 days of drought treatment, the highest and lowest OP values were observed in Osmancik-97 (-1.14 MPa) and Karacadag (-1.55 MPa) varieties, respectively. In the same treatment, it was observed that the amount of proline increased 19.9-fold in Karacadag and 3.6-fold in Osmancik-97. When the data obtained from all parameters were correlated with drought stress tolerance, Osmancik-97 and Beser varieties were considered to be tolerant, while Gonen and Karacadag varieties are considered to be more sensitive.Öğe The Evaluation of Total Phenolic, Flavonoid, Sugar Contents and Antioxidant Activity of Tayfi Grape in Turkey(Batman Üniversitesi, 2017) Kuru, İbrahim Selçuk; Orcan, Pınar Karakuş; Akbaş, Filiz; Işıkalan, Çiğdem; Namlı, SüreyyaTayfi (Vitis vinifera L.) only grown in the Southeastern Anatolia Region provides a major contribution on the economy of the region people. But, the research related to nutritional content of this species was not found. This study was carried out to determine the total phenolic, flavonoids, sugar content and antioxidant activity of different organe (mature/young seed and leaf ) ‘Tayfi’ grape. The highest amount of total phenolic (380,94 µg GAE/mg extract) was found to be in the young seeds. The high amount of total flavonoid content (107,21 µg QEs/mg extract) was obtained from young leaves. Higher antioxidant activity was observed in young seeds (91,32%) as expected from high contents of total phenolics. In general, as the total phenolics in seed increased, the antioxidant activities also increased. Also, changes in sugar content (glucose, fructose, sucrose and maltose) were determined in leaf from seed (young and mature) of Tayfi grape by HPLC. The highest concentration of sucrose was descripted in mature seed (9,29 mg/g).
