Türkiye’de yetişen endemik (Nepeta nuda subsp.lydiae) bitkisine ait farklı ekstrelerin antioksidan aktivitelerinin belirlenmesi ve fenolik bileşik içeriklerinin LC-MS/MS ile analizi

Özet
Nepeta nuda subsp. lydiae bitkisinin antioksidan aktivitelerini belirlemek için ABTS (2,2´-azinobis 3-etilbenztiyoazolin-6-sülfonik asit) radikal giderme aktivitesi, DPPH (1,1-difenil-2-pikril-hidrazil) serbest radikal giderme aktivitesi, FRAP (ferrik iyonları indirgeme gücü) ve CUPRAC (kuprik iyonları indirgeme kapasitesi) biyoanalitik metotları in vitro olarak çalışıldı. Antioksidan çalışmalarda sonuçlar standart antioksidanlar BHA, BHT ve askorbik asit ile karşılaştırıldı. Bu çalışmada Nepeta nuda subsp. lydiae bitkisinin liyofilize su ekstresinin DPPH radikal giderme aktivitesi ve kuprik (Cu2+) iyonlarını indirgeme kuvveti orta düzeyde, ABTS radikal giderme aktiviteleri ile ferrik (Fe3+) indirgeme kuvveti ise düşük seviyede aktivite gösterdiği tespit edildi. Bununla beraber ekstrelerin antimikrobiyal aktiviteleri disk difüzyon yöntemi ile belirlendi. Bu çalışmada, Nepeta nuda subsp. lydiae ekstresi B.megaterium'a karşı en yüksek anti-bakteriyel aktivite sergilerken en düşük etkisini P. aeruginosa ve K. pneumoniae'ye karşı sergilemiştir. C.albicans'a karşı yüksek antifungal aktivite gösterirken Y. lipolytica ve S. cereviciae'a karşı herhangi bir etki göstermemiştir. LC-MS/MS analiz metodu ile Nepeta nuda subsp. lydiae bitkisinin fenolik içerik analizi yapıldı. Kuinik asit, klorojenik asit, rosmarinik asit ve kumarik asit en çok bulunan fenolik bileşikler oldu. Kafeik asit, kaempferol, luteolin, apigenin, rhamnetin ve krisin fenolik asit miktarları nisbeten daha az olduğu belirlendi. Sonuç olarak Nepeta. nuda subsp. lydiae ekstreleri yüksek miktarda fenolik bileşik içermeleri biyolojik faaliyetler ile ilişkilendirilebilir. The antioxidant activities of ethanol and water extracts of Nepeta nuda subsp. lydiae leaves were determined by different in vitro methods such as; DPPH, ABTS, FRAP and CUPRAC assays. The results were compared to BHA, BHT and ascorbic acid as standard antioxidant compounds. Both water and ethanol extracts presented moderate antioxidant potential on DPPH free radical scavenging assay and CUPRAC assay. Also, both extracts presented low level on ABTS and FRAP assays. Also, the antimicrobial activity of Nepeta nuda subsp. lydiae was determinated by the disc diffusion method. In this study, the extract of Nepata nuda subsp. lydiae was showed the highest antibacterial activity against the B. megaterium. At the same time the lowest antibacterial activity was showed against P. aeruginosa and K. pneumoniae. The extract of Nepata nuda subsp. lydiae showed the highest antifungal activity against C. albicans but it did not show any significant antifungal activity against Y. lipolytica and S. cereviciae. In another step of this study, phenolic compounds of Nepeta nuda subsp. lydiae leaves were identified by LC-MS/MS. Chlorogenic acid, rosmarinic acid, coumaric acid and quinic acid, were found to be the most abundant compounds in the methanol extract of N. nuda subsp. lydiae, among the twenty-seven compounds studied by LC–MS/MS. Also, smaller amounts of kaempferol, p-coumaric acid, tr-caffeic acid, apigenin, luteolin and rhamnetin were identified, quantitatively. In conclusion, leaves of N. nuda subsp. lydiae have high potential of phenolic contents that mainly attributed with biological activities.
