Özet
Bu çalışmada sıcak su kaynaklarından izole edilen bakterilere ait enzimlerin endüstriyel alanda önem arz etmelerinden dolayı Bingöl İli Ilıcalar beldesinden getirilen su örneklerinden bakteriler izole edilmiştir. İzole edilen bu bakterilerin endüstriyel açıdan önemli olan enzimleri üretme durumları araştırılıp bu enzimler üzerine çalışmalar yapılması amaçlanmıştır. Yapılan bu çalışmada izole edilen su ve toprak örneklerinde 2 bakteri izole edildi. Bu bakterilerin çeşitli biyokimyasal testleri ile morfolojik ve fizyolojik analizleri yapıldı. İzole edilen 4NK bakterisinin çubuk şeklinde gram pozitif olduğu, 5NK bakterisinin de gram pozitif ve çubuk şeklinde olduğu tespiti yapılmıştır.İzole edilmiş bu iki bakteri için optimum pH değerinin 6 olduğu tespit edildi.Optimum üreme sıcaklıkları ise sırasıyla 40 ve 45 oC olarak belirlenmiştir. İzole edilen bakterilerin optimum koşulları belirlendikten sonra bunların ürettikleri enzimlerin optimumları belirlenmeye çalışılmıştır. Buna göre 4NK bakterisinin proteaz ve amilaz enzimlerini üretim açısından optimum süreleri sırasıyla 24. ve 15. saatler olduğu belirlenmiştir. Proteaz ve amilaz enzimlerinin optimum pH’ları sırasıyla 8 ve 4-11; optimum sıcaklıkları sırasıyla 40 ve 60 oC olarak bulunmuştur. 5NK bakterisinin ise proteaz ve amilaz enzimlerini üretim açısından optimum süreleri sırasıyla 24. ve 12.saat olarak belirlenmiştir. Bu bakteri varyetesinin proteaz ve amilaz enzimlerinin optimum pH’ları 8 ve 4-11; optimum sıcaklıkları 50 oC olarak bulunmuştur. İzolasyonu yapılan 4NK ve 5NK bakterilerinin 16S rRNA dizi analizi yapılmış, buna göre filogenetik olarak hangi tür ve alt türlere yakın olduğu tespiti yapılmıştır. Analiz sonucu 4NK bakterisinin Bacillus subtilis türüne yakın, 5NK bakterisinin ise Bacillus paralicheniformis türüne filogenetik olarak yakın olduğu tespit edilmiştir. In this study, bacterial isolates were isolated from the Bingöl Ilicalar landfill samples due to the importance of bacterial enzymes isolated from hot water sources in the industrial area. It is aimed to investigate the production of industrially important enzymes by these isolated bacteria and to work on these enzymes. In this study, 2 bacteria were isolated in isolated water and soil specimens. Various biochemical tests, morphological and physiological analyzes of these bacteria were performed. It was determined that the isolated 4NK bacteria were rod-shaped gram positive and 5NK bacteria were gram positive and rod-shaped. The optimum pH value for these two isolated bacteria was determined to be 6. Optimum reproductive temperatures were 40 and 45 oC, respectively. Once the optimum conditions of the isolated bacteria have been determined, attempts have been made to determine the optimums of the enzymes they produce. Accordingly, it has been determined that the optimal time for production of protease and amylase enzymes of 4NK bacteria is 24th and 15th hours, respectively. The optimum pHs of protease and amylase enzymes were 8 and 4-11, respectively; optimum temperatures were found to be 40 and 60 oC, respectively. The optimal time for production of protease and amylase enzymes of 5NK bacteria was determined to be 24th and 12th hours, respectively. The optimum pHs of protease and amylase enzymes of this bacterial variant were 8 and 4-11; the optimum temperatures were found to be 50 oC. 16S rRNA sequence analysis of isolated 4NK and 5NK bacteria was performed and it was determined which species and subspecies are phylogenetically close to each other. It was determined that the resultant 4NK bacteria were close to Bacillus subtilis and 5NK bacteria were phylogenetically close to Bacillus paralicheniformis.
