    Determination of Antioxidant, Antidiabetic, Anticholinergic, Antiglaucoma Properties and Comprehensive Phytochemical Content by LC-MS/MS of Bingöl Honeybee Pollen

    Honeybee pollen is the most important food and protein source for bees. It is a highly nutritious natural product for humans due to its content. Pollen shows different phytochemical content and variable biological activity according to different geography and vegetation. Therefore, in this study, the comprehensive phytochemical content by LC-MS/MS, and its antioxidant properties by different assays, with enzyme inhibition potential; antidiabetic, anticholinergic, and antiglaucoma properties, were determined from a pollen sample from Bingö l province, one of the significant beekeeping centers in Tü rkiye. As a result of LC-MS/MS, 15 metabolites were determined, and the highest concentration of quinic acid (1.531 mg analyte/g extract) was found. Antioxidant results: total phenolic content 113.14 mg GAE/g, total flavonoid content 64.11 mg QE/g, Fe3+ reduction 0.43 mu g/mL, CUPRAC 0.511 mu g/mL, FRAP 0.976 mu g/mL, DPPH IC50: 1.06 mu g/mL, ABTS IC50: 0.933 mu g/mL, and DMPD IC50: 0.598 mu g/mL. In addition, pollen showed antiglaucoma, anticholinergic, and antidiabetic properties by inhibiting carbonic anhydrase isoenzyme II (hCA II), acetylcholinesterase (AChE), and alpha- amylase enzymes, respectively. In this study, it was determined that Bingö l pollen has a comprehensive phytochemical content and antioxidant properties and, for the first time, inhibits hCA II, AChE, and alpha-amylase enzymes.

    DC-link voltage stability enhancement in intermittent microgrids using coordinated reserve energy management strategy

    In recent years, due to its cost effectiveness and environmental advantages, demand for renewable energy resources has grown and their contributions to grid power has therefore increased while requiring effective frequency and voltage regulation. DC link voltage instability is a potential problem in solar energy microgrids, especially during an intermittency, where the system reliability degrades and DC link capacitor is under higher stress. In this article, a novel reserve energy management scheme based on battery and super capacitor storage is presented to stabilize the DC link voltage and reduce capacitor stress, while enhancing the system reliability. The scheme is tested in four different scenarios: Inverter connected DC-microgrid with irradiance intermittencies, standalone DC-microgrid without inverter and irradiance intermittencies, standalone DC-microgrid without inverter and load intermittencies, and standalone DC-microgrid with inverter under irradiance intermittencies. Simulation results indicate that the proposed control strategy stabilizes DC link voltage over all scenarios, even subject to large instances of irradiance or load changes. During low solar irradiance, the battery and super-capacitor promote voltage stability by compensating power deficits from the utility grid in the inverter connected grid case. In stand alone mode, the battery provides power during intermittencies and the supercapacitor provides fast transient voltage compensation. The strategy is notable in reducing stress on DClink capacitors and mitigating inverter voltage fluctuations, ultimately enhancing inverter longevity. The results show that the proposed control scheme can improve voltage stability, mitigate the transient effects and guarantee the reliable operation of solar microgrids in variable conditions.

    Blood-based biomarkers of frailty in older patients with cancer

    Purpose of reviewThis review aims to evaluate the current evidence on blood-based biomarkers for frailty detection in older cancer patients. It explores the potential of various biomarkers, including inflammatory markers and microRNAs (miRNAs), to serve as indicators of frailty and examines the limitations of existing studies. The review also highlights the need for further research to validate these biomarkers and improve their clinical applicability.Recent findingsRecent studies have examined blood biomarkers associated with frailty in older cancer patients. Findings suggest that elevated granulocyte levels and lower macrophage-derived stem cells and regulatory T cells are linked to frailty. Inflammatory biomarkers such as interleukin-6 and specific miRNAs, as well as higher neutrophil-to-lymphocyte ratios, have also been identified as potential indicators of frailty. While these biomarkers show promise, no single marker has proven sufficient, and combining them may improve frailty detection. Further research is needed to validate their clinical usefulness in this population.SummaryBlood-based biomarkers show potential for detecting frailty in older patients with cancer, but further research is needed, particularly beyond an inflammatory focus and with more robust study designs.

    Design, synthesis, structural characterization, in vitro anticancer activity, and in silico studies of some new hydroxylated and fluorinated-substituted hydrazone derivatives

    Today, due to improved lifestyles and increased survival, the number of new cancer cases and cancer-related deaths continues to increase. In this study, novel hydroxylated and fluorinated-substituted hydrazone derivatives bearing an aromatic nitro moiety (2a-d and 3a-d) were designed as potential anticancer drug candidates, synthesized for the first time, and evaluated for their anticancer activity against chondrosarcoma (SW1353), a common primary malignant cartilage-forming tumor, neuroblastoma (SH-SY5Y), a type of brain cancer, and healthy (L929) cell lines using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) method for 24 h. The chemical structures of the target molecules were confirmed by FT-IR, 1D NMR (1Hand 13C- NMR, and APT), 2D NMR (COSY, HETCOR, and HMBC), and elemental analysis. Some of the compounds targeted against these cancer cell lines showed activity greater than 200 mu M, whereas others (2d and 3a) demonstrated significant cytotoxic activity. Among them, compound 3a (IC50 = 9.45 +/- 2.14 mu M), a fluorinatedsubstituted hydrazone derivative, showed significant cytotoxic activity against the human SW1353 cell line compared to cisplatin (IC50 = 11.9 +/- 0.95 mu M). The anti-migratory properties of compounds 2d and 3a in SW1353 cells, were investigated. In particular, compound 3a exhibited anti-migration behavior in SW1353 cells, with a wound closure rate of 22.25 % compared with control cells. Further, scaffolds 2d and 3a exhibited the best docking with target receptor proteins 2OH4 (-8.3 and -8.4 kcal/mol) and 3QX3 (-12.2 and 11.0 kcal/mol), thereby supporting our bioactivity studies. Compounds 2a, 3a, 3b, and 3c showed high gastrointestinal (GI) absorption, with all except 3a being non-permeable to the blood-brain barrier (BBB). Most compounds, except 3d, are non-substrates of P-glycoprotein (P-gp). In conclusion, the in vitro and in silico results of some of the tested compounds indicate that they could be promising molecular frameworks for further studies.

    Wound healing activity of Salvia huberi ethanolic extract in streptozocininduced diabetic rats

    Objective: The aim of this study was to examine the in vivo wound healing potential of Salvia huberi Hedge (endemic to Turkey) on excision and incision wound models in diabetic rats. Method: Male Wistar albino rats, 3-4 months old and weighing 180-240g were used. The animals were randomly divided into five groups including Control, Vehicle and Fito reference, and two different concentrations (0.5% and 1% weight/weight (w/w)) of ethanol extract of Salvia huberi were investigated in both wound models on streptozocin-induced diabetic rats using macroscopic, biomechanical, biochemical, histopathological, genotoxic and gene expression methods over both seven and 14 days. Fito cream (Tripharma Drug Industry and Trade Inc., Turkey) was used as the reference drug. Results: A total of 60 rats were used in this study. Salvia huberi ointments at 0.5% and 1% (w/w) concentrations and Fito cream showed 99.3%, 99.4% and 99.1% contraction for excision wounds, and 99.9%, 97.0% and 99% contraction for incision wounds, respectively. In Salvia huberi ointments and Fito cream groups, re-epithelialisation increased dramatically by both day 7 and day 14 (p<0.05). By day 14, low hydroxyproline and malondialdehyde (MDA) levels, and high glutathione (GSH) levels were observed in the Salvia huberi ointment groups. After two application periods, damaged cell percent and genetic damage index values and micronucleus frequency of Salvia huberi ointment treatment groups were lower than Control and Vehicle groups (p<0.001). A growth factor expression reached a high level by day 7 in the Control group; in Salvia huberi-treated groups it was decreased. Conclusion: The study showed that application of Salvia huberi ointments ameliorated the healing process in diabetic rats with excisional and incisional wounds and may serve as a potent healing agent.     Heavy Metal Levels and Risk Assessment of Walnut Genotypes ( Juglans regia L.) Grown in Aras Basin of Turkiye

    Walnuts are among of the most popular nuts worldwide due to their health-promoting compounds, and their consumption is on the rise. This increase in walnut consumption may lead to an increase in the intake of heavy metals in the body. In this study, the levels of the heavy metals Cr, Ni, Pb and Cd were determined in a total of twenty walnut genotypes grown in four different locations in the Aras Basin, and the distribution of these heavy metals according to location was analyzed. While highly significant differences (P<0.001) were found among the walnut genotypes in terms of heavy metal contents, the distribution of these metals among the sites showed differences (P <= 0.05). The average amounts of Cr, Ni, Cd and Pb in the walnuts were 476.00 mu g kg(-1), 202.83 mu g kg(-1), 2.06 mu g kg(-1) and 30.64 mu g kg(-1), respectively. All heavy metals analyzed according to location showed the highest values in Tuzluca. As a result, since the hazard quotient (HQ) which derived from the daily intake rate (DIR) of each trace element analyzed was less than 1(HQ<1), it was found that the investigated walnut genotypes were not risky for human health.

    Presynchronization in Sheep Ensures Synchronization of Next Estrus and Improves Fertility

    In this study, fertility was evaluated either in the first hormonally synchronized or in the subsequent natural estrus among 202 sheep during the breeding season. Intramuscular (im) PGF(2 alpha )was administered 11 days apart in both the PG (n=50) and Pre-PG (n=50) groups, while im PGF(2 alpha )was given at the removal of an intravaginal progesterone sponge (after 11 days) in the Sponge (n=51) and Pre-Sponge (n=51) groups. Estrus was monitored following the last hormonal application (day 0) in all groups and starting from day 16 in the Pre-PG and Pre-Sponge groups for 5 days. Ewes were mated during the first hormonally synchronized estrus (PG and Sponge groups) or in the next (natural) estrus (Pre-PG and Pre-Sponge groups). The estrus rate and litter size in the Pre-Sponge group were higher than those in the other groups (P<0.05). In the Pre-Sponge group, pregnancy and lambing rates were higher than in the PG and Sponge groups, and fecundity was higher than in the PG group (P<0.05). It was concluded that progesterone and PGF(2 alpha )-based estrus synchronization may negatively affect fertility. Additionally, the next estrus is also synchronized after using progesterone and PGF(2 alpha )-based synchronization protocols. Fertility loss caused by the application of exogenous hormones can be mitigated by mating at the next natural estrus. Furthermore, the next natural estrus can be utilized in situations where hormone use is inappropriate but synchronization is desired.

    Retrospective analysis of metabolic effects of laparoscopic gastric plication and single anastomosis duodeno-ileal bypass with sleeve gastrectomy (SADI-S)

    AIM: The aim of this study is to investigate the clinical outcomes of patients with type 2 diabetes mellitus (T2DM) who underwent laparoscopic gastric plication (LGP) or single anastomosis duodeno-ileal bypass with sleeve gastrectomy (SADIS) and analyze the postoperative first-year results of these two operations. METHODS: Forty-three patients who have undergone LGP and 36 patients who had undergone SADI-S were included in this study. Baseline and postoperative first-year data of patients with T2DM who have undergone LGP or SADI-S in our clinic between January 1, 2013, and June 30, 2016, were retrospectively analyzed. RESULTS: It is understood that both operations maintained a remarkable improvement in blood glucose parameters alongside total cholesterol and triglyceride levels. The complete diabetes remission rate was significantly higher in the SADI-S group than in the LGP group (69.4% vs. 42.1%, p=0.018). LGP group achieved better results than SADI-S on weight loss in terms of the percentage excess weight loss (EWL%) (p<0.001) and the percentage total weight loss (TWL%) (p<0.001). CONCLUSION: In our study, both operations facilitated diabetes remission, and complete remission or improvement was obtained in most of the cases. In addition, statistically significant weight loss was observed in both procedures. Therefore, both bariatric techniques can be chosen for obese diabetic patients considering their priorities and needs.

    Sensor properties of plasmonic silver and gold nanoparticles produced by pulsed laser deposition

    In this study, 20-100 nm Au and Ag noble metal nanoparticles were produced using the Pulse Laser Deposition technique, and very sensitive optical properties of nanoparticles were investigated using the LSPR technique. The morphology of the produced nanoparticles was determined and characterized using scanning electron microscopy (SEM). The chemical bond structure of the Biotin molecule was investigated by Fourier Transform Infrared Spectroscopy (FTIR). In addition, the UV-Vis spectroscopy technique was used to determine LSPR absorption peaks of nanoparticles. Au and Ag Nanoparticles were deposited on the glass substrate depending on the laser deposition times. As the laser deposition time of the produced nanoparticles increased, both the size of the formed nanoparticles and the number of nanoparticles per unit surface increased. It was observed that the shape of Au and Ag nanoparticles produced based on SEM images was spherical. When UV-Vis spectra of Au and Ag nanoparticles were examined, it was observed that LSPR peaks shifted to longer wavelengths (redshift) as the laser deposition times increased. We have observed, to our knowledge, that the highest LSPR peak appeared at 1065 nm in the near-infrared region for plasmonic Au nanoparticles produced by Pulsed Laser Deposition. Once LSPR peaks of the plasmonic nanoparticles were characterized, Biotin molecules with different concentrations were attached to Ag and Au NPs to detect the sensor properties. LSPR peak shifts of bound nanoparticles could be observed and a blueshift of peaks was demonstrated as the concentration increased.

    Observations on the breeding ecology of Lesser Kestrel Falco naumanni (Fleischer 1818) at a new breeding site in southeastern Turkiye

    A new breeding site for Lesser Kestrel was identified and the nesting area was monitored between 2019- 2022 at Diyarbakir, Turkiye. Birds arrive at the nesting site at the end of March and breeding commences with courtship behaviour and copulation at the beginning of April. Incubation starts in mid April and lasts until mid-May when the eggs hatch. The juveniles fledge at the end of June, and begin to practice hunting with adults in early July. Further monitoring of the population is recommended.

    A different perspective on the treatment of branchial cleft cysts, which are very rarely located in the nasopharynx: a case series

    Nasopharyngeal branchial cleft cysts are very rare. There is no consensus about the approach to these pathologies. The problem that makes surgery difficult is its possible proximity to vital vascular structures such as internal carotid in the deep plane and located laterally. We aimed to present our series of three patients who were diagnosed with nasopharyngeal branchial cyst and operated by transnasal endoscopic method by determining the safe surgical margin with the help of Doppler ultrasonography. Three male patients, who had various complaints such as difficulty breathing through the nose and sleep problems, were operated on with a transnasal endoscopic approach supported by 20MHz Doppler ultrasonography due to cystic formation in the nasopharynx. Histopathological evaluations were reported as a nasopharyngeal branchial cleft cyst. Doppler ultrasonography assisted transnasal endoscopic approach can be applied in the treatment. This approach both shortens the duration of surgery and minimizes the risk of neurovascular injury.
